Objective:The purpose of this study is to detect the trend of peripheral blood lymphocyte subsets, through the chemotherapy before, during and after in malignant tumor radiotherapy. Explore the correlation between the peripheral blood lymphocyte subsets and radiation therapy of malignant tumors. Applied high-energy carbon ion beams (12C6+) level fixed beam, explore the influence of carbon ion irradiation on lymphocyte subsets in patients with radiation therapy and the relationship between sensitivity and immune function.Method:1. Using FACS Calibur flow cytometric to test peripheral T lymphocyte subsets for46patients with head and neck squamous cell carcinoma.2. Applied FACSort flow cytometer to detect the peripheral blood lymphocyte subsets of61cases of squamous cell carcinoma, to measured lymphocyte subsets indicators for each patient one time before radiotherapy, during and after, and compared the test results.3. Several T cell subsets (T, Th, Ts, NK)and the CD4+/CD8+ratio in peripheral blood lymphocytes from46breast cancer patients were analyzed by standard four-color flow cytometry on a BD FACS Calibur. Immunohistochemistry was used to assess expression levels of ER, PR, and Her-2in breast cancer tissue. The correlation between T cell subsets and expression of ER, PR, and Her-2in breast cancer patients was analyzed before and after postoperative radiotherapy.4.23Patients with malignant tumor received carbon ion beam radiation therapy from2010to2011,and detected the T-cell subsets with flow cytometry at the first day、six day and after radiation therapy,then compared their test values.Results:1. The head and neck squamous cell carcinoma patients before, during and after the treatment CD3+, CD4+, CD8+, NK cells had no obvious changes, B cells and CD4+/CD8+ratio after radiotherapy changed significantly(P<0.05).2. The value of before radiotherapy and radiotherapy of the moderately differentiated squamous cell carcinoma patients:CD3+, B lymphocytes the difference was significant (P<0.05); before radiotherapy and after radiotherapy: the difference of CD3+, CD8+, CD4+, B lymphocytes, CD4+/CD8+levels were statistically significant (P<0.05.); Radiotherapy and after radiotherapy:the difference of CD8+, CD4+, B lymphocytes, CD4+/CD8+levels were significant (P<0.05.). The correlation analysis between the differences in the difference group and the efficacy of radiotherapy:the difference of B lymphocytes after radiotherapy and the efficacy of radiotherapy were negative correlation (r=-0.343, P=0.007), the difference of CD8+before and after radiotherapy and radiotherapy efficacy was positively correlated (r=0.331, P=0.009), the difference between the remaining difference and the radiotherapy group was no significant (P>0.05). There are differences in the difference between the treatment groups after radiotherapy with no significant difference between correlation (P>0.05).3. Expression of the ER receptor and PR receptor, which strongly correlated with Th cells (P=0.000and0.014, respectively), was significantly higher in the Th subsets that changed significantly as a result of radiotherapy than in the Th subsets that did not change significantly.No correlation was observed between T cell subsets and expression of ER, PR, or Her-2before and after postoperative radiotherapy (P>0.05).4. The total mature T cells decreased significantly after radiotherapy than before(P=0.002).The variation of Ts cells at the first day、six day and after radiation therapy were statistical significant difference (P=0.003and0.001). A trend downward of the Th cells and the ratio of CD4+/CD8+were observed during radiotherapy, but there were no significant difference.The level of NK and B cells after radiation therapy were less than before radiotherapy’s, there were no significant difference either(P=0.390and0.701).Conclusion:1. To test the immune function of patients with head and neck squamous cell carcinoma after treatment is useful.2. Moderately differentiated squamous cell carcinoma patients before radiotherapy, during and after peripheral blood lymphocyte subsets in patients with cell free tips Epidemic and humoral immune function disorder in varying degrees, B lymphocytes and CD8+cells and the efficacy of radiotherapy in patients with mid-correlated radiation effects on immune function, immune function to explore the relevance of long-term efficacy of radiotherapy needs further research.3. Patients who are ER or PR negative may show decreased immune function and poor prognosis after treatment with postoperative.4. The carbon ion beam radiation therapy could injure the cellular immune function to a certain degree.
