Objective To investigate the relationship between sphingosine-1-phosphate receptors and expression of matrixmetalloprotenases-9(MMP-9) and restenosis, pathological morphology of injured blood vessel and expression of S1P1, S1P2, Rac-1and MMP-9were observed through HE stain, Real-time PCR and Western blot.Method80Wistar rats were randomly divided into sham group (A group), balloon injury group (group B) and balloon injury+FTY720group (group C), each group was divided into seven days,14days,21days,28days four subgroups. Morphology of the vessel wall hyperplasia was observed by light microscopy. We also use real-time PCR to assay S1P1and S1P2, and Western blot to assay expression of Rac-land MMP-9.Result No significant changes has been found when we investigate all indicatrix in group A. Whereas expression of each index in group B7days after injury significantly increased, then gradually reduced. Comparing with group A at each time nodes factors that we observed was so different(P<0.05). the test result of each factor in group C were lower than in group B but higher than in group A (P<0.05). Morphological result showed vessel wall in group B was thicker comparing with in group A, and it gets more significantly as time goes by. And each time node was observed in group C that showed the extent of vessel wall thickening was significantly reduced compared with in group B.Conclusion1. FTY720can significantly reduce the rat carotid artery stenosis after balloon injury.2. TY720can reduce the expression of MMP-9after vascular injury.
