| Objective Hepatocellular carcinoma (Hepatocellular Carcinoma, HCC) isone of the most common malignant tumor, the first leading cause of death due tocancer, but its carcinogenesis is still in the study. Through the research of NQO1,P53and Survivin three gene expression of mRNA and protein in hepatocellularcarcinoma tissues in different pathological gradings, to investigate therelationship between the three genes and hepatocellular carcinoma.Methods Cells were cultured with RT-PCR NQO1and Survivin mRNAanalysis showed the four strains of HCC cell line with various degrees ofdifferentiation; collection of pathological wax block and intraoperative frozenspecimen sections, pleomorphic way using RFLP to identify NQO1, usingimmunohistochemical staining and Western blot method to explore the geneexpression of P53and survivin protein in hepatocellular carcinoma tissues indifferent pathological gradings.Results NQO1mRNA in HCC tissues compared with normal tissuesrelatively high in expression of NQO1in patients with hepatocellular carcinoma, and moderately and low differentiated mutation probability compared with thehigh differentiated of liver cancer patients, there was increasing trend (p<0.05);the proportion of polymorphic NQO1to mutant type in HCC tissues in thepathological grading of: high differentiated-9.1%, moderately differentiated–38.7%, low differentiated-40.0%, NQO1mutation probability in the moderatelyand third low differentiated of HCC patients, there were significant increased(p<0.05); P53protein expression of mutant form in different groups are similar,and more than50%, So the mutant form P53correlates with the occurrence ofHCC, and no significant association with hepatocellular carcinoma grading;Survivin mRNA expression of Survivin in hepatocellular carcinoma cells than innormal tissue of the control group (T/N ratio>1) high, but the T/Nratio in eachgroup had no significant difference; Survivin protein expression in tissuesections of each HCC grading and expression in peripheral normal liver cellcytoplasm, there is manifestation of survivin in hepatocellular carcinoma cells,and found that survivin exists in the cytoplasm but is less than in normal tissue,a few samples found that survivin exists in the cell Nucleus; Survivin expressionin hepatocellular carcinoma cells the content of Survivin mRNA in fourhepatoma cells had no obvious difference.Conclusion Overexpression of P53gene mutation, NQO1polymorphism,NQO1and Survivin mRNA play an important role in the process ofhepatocellular carcinoma, while the Survivin isoforms is involved in the processof hepatocarcinogenesis is discussed. |
