| Background and purpose: The single nucleotide polymorphism (SNP) ofrs7395662in the MADD-FOLH1has been associated with serum lipid traits, butthe results are inconsistent in different populations. The present study wasundertaken to investigate the association of MADD-FOLH1rs7395662SNP andseveral environmental factors with serum lipid levels in the Guangxi Mulao andHan populations.Method: A total of721subjects of Mulao and727participants of HanChinese were randomly selected from our previous stratified (sex and age)randomized samples. Genotyping of the MADD-FOLH1rs7395662SNP wasperformed by polymerase chain reaction and restriction fragment lengthpolymorphism combined with gel electrophoreóis, and then confirmed by directsequencing.Results: Serum apolipoprotein (Apo) B levels were higher in Mulao than inHan (P <0.01). The allelic and genotypic freq encies in Han were differentbetween males and females (P <0.05), but there was no significant differencebetween Mulao and Han or between Mulao males and females. The levels oflow-density lipoprotein cholesterol (LDL-C) and ApoB in Mulao females were different among the genotypes (P <0.05), the G allele carriers had higherLDL-C and ApoB levels than the G allele non-carriers. The levels of totalcholesterol (TC), triglyceride (TG), LDL-C and ApoB in Han males and TC, TGand high-density lipoprotein cholesterol (HDL-C) in Han females were differentamong the genotypes (P <0.05-0.01), the subjects with GG genotype in Hanmales had higher TC, TG, and ApoB and lower LDL-C levels than the subjectswith AA or AG genotype, and the G allele carriers in Han females had lower TCand HDL-C levels than the G allele non-carriers. The levels of LDL-C andApoB in Mulao females were correlated with the genotypes (P <0.05for each).The levels of HDL-C and ApoAI in Han males and HDL-C in Han females werecorrelated with genotypes (P <0.05-0.001). Serum lipid parameters were alsocorrelated with several environmental factors such as age, sex, body mass index,alcohol consumption, cigarette smoking and blood pressure in both ethnicgroups (P <0.05-0.01).Conclusion: The association of MADD-FOLH1rs7395662SNP and serumlipid levels is different between the Mulao and Han populations, and betweenmales and females in both ethnic groups. There may be an ethnic-and/orsex-specific association of the MADD-FOLH1rs7395662SNP and serum lipidparameters in our study populations. |
