Objective: To disscus the clinical application of biological markers smallbreast epithelial mucin,human mammaglobin, and cytokeratin19by detectingcirculating tumor cells and lymph node micrometastasis in breast cancer.Methods: The peripheral blood and tissue samples were collect in diagnosedpatients of breast neoplasms from April13,2012to October10,2012,Department of Breast Surgery,Tumor Hospital Affiliated to Guangxi MedicalUniversity. SBEM, hMAM, and CK19mRNA were dected by nested PCR inperipheral blood of47cases of breast cancer and45cases of benign breasttumor. RT-PCR method was used to detected the mRNA express of10cases ofbreast cancer tissue,10cases of paracancerous tissue and10cases of benignbreast tumor tissue. SBEM, hMAM, and CK19were detected by usingimmunohistochemistry technique in27cases breast cancer patients’109lymphnodes.Results: SBEM expression in peripheral blood of breast cancer and benignbreast tumor patients were82.98%and62.22%,P=0.02;h MAM expression in peripheral blood of breast cancer and benign breast tumor patients were65.96%and42.22%, P=0.02; CK19expression in peripheral blood of breast cancer andbenign breast tumor patients were89.36%and80.00%,P=0.21.The52lymphnodes metastasis by HE staining strongly. Detecting by CK19, there have5lymph nodes micometastasis in57negative lymph nodes. hMAM and SBEMexpression in metastatic lymph nodes is weaker than that of CK19, and thepositive cells were not uniform.Conclusions: SBEM, hMAM, and CK19as biomarkers for breast cancermicrometastases in peripheral blood have high sensitivity and low specificity.Joint detection could improve the specificity and it has some clinicalsignificance for the early detection of breast cancer micrometastases.CK19maybe used to detect axillary lymph node micrometastases.
