Effect Of Peripheral Blood Mononuclear Cells DNA Methyltransferase And CD11a Gene Expression On Anti-neutrophil Cytoplasmic Antibodies-associated Vascmlitis

Objective：To investigate the relationship of peripheral blood mononuclearcells (PBMC) DNA methyltransferase1(DNMT1), methyltransferase3A(DNMT3A), methyltransferase3B(DNMT3B) and CD11a mRNA expressionin primary ANCA-associated small vasculitis(AAV).Methods：The mRNA level of three kinds of DNMTs and CD11a inperipheral blood mononuclear cells(PBMC) from33cases of active phrase ofAAV were examined and60healthy controls was adopted, it was contrasted byReal-time fluorescent quantitation reverse transcription polymerase chainreaction（RT-qPCR）,the expression of differences genes compared betweenAAV groups and healthy controls. The vital signs, serum biochemicalparameters and immune indexes were detected.Results:1、 Significant statistics difference was found in the mRNAexpression of DNMT1between AAV group and healthy controls (P＜0.01).Theexpression of DNMT1in PBMC was more decreased than that in healthy controls；Significant statistics difference was found in the mRNA expression ofCD11a between AAV group and healthy controls (P＜0.05)；No significantstatistics difference was found in the mRNA expression of DNMT3A andDNMT3B between AAV group and healthy controls(P＞0.05)；The mRNAexpression of CD11a in PBMC were higher than healthy controls；The mRNAexpression levels of DNMT1in PBMC of AAV patients and BVAS scoreshowed a negative correlation（P＜0.05)；The mRNA expression levels ofCD11a in PBMC of AAV patients and BVAS score were contrasted a positivecorrelation(P＜0.05)；The mRNA expression levels of DNMT1and CD11awere contrasted a negative correlation（P＜0.05)；2、The levels of total cholesterol，triglyceride，blood urea nitrogen，serumcreatinine，blood uric acid，C-reactive protein，blood sedimentation, white bloodcell count in AAV group were significantly higher than control group(P＜0.05)；While，hemoglobin，serum albumin levels were lower than the healthycontrols(P＜0.05)；However, No significant statistics difference was found inblood glucose (P＞0.05).Conclusions：1、The mRNA expression of DNMT1present in PBMC inpatients with primary AAV was decreased；2、Increased mRNA expression ofCD11a causative genes was found in PBMC in patients with primary AAV；3、The mRNA expression levels of DNMT1and CD11a genes may negativelycorrelated.
