AIM:To observe the expression of Smurf1,Smurf2,Smad3and Smad7protein in liver fibrosis tissues of human and to evaluate their interaction and themechanism of signals in liver fibrosis.METHODS:Immunohistochemistry was applied to detect the expression ofSmurf1,Smurf2,Smad3and Smad7in9tissues of normal liver and38tissues ofchronic HBV infections.RESULTS:Smurf1,Smurf2,Smad3and Smad7showed widespreadexpression.in the liver parenchymal cells and nonparenchymal cells. Comparingwith normal liver, the expression of Smad3increased significantly(Z=-3.110,P=0.002), the expression of Smurf2also increased significantly (Z=-2.716,P=0.007),the expression of Smad7decreased significantly in iver fibrosisgroup(Z=-2.899,P=0.004);the expression of Smurf1had no significant change(Z=-1.399,P=0.162),it showed no significance.There was significant positivecorrelation between Smad3and the degree of fibrosis (r=0.627,P=0.000);Smad7was negatively correlated with the degree of fibrosis(r=-0.488,P=0.000);There was no significant correlation between Smurf1and liver fibrosis (r=-0.064,P=0.670); There was significant positivecorrelation between Smurf2and the degree of fibrosis r=0.652, P=0.000).Smurf2was positively correlated with Smad3r=0.523,P=0.000)and negatively correlated with Smad7(r=-0.447,P=0.002);There was norelationship between Smurf1and Smurf2, Smad3,Smad7(r=-0.008、r=-0.053、r=0.219,all P>0.05);There was a negative correlation between Smad3andSmad7(r=-0.389,P=0.007).CONCLUSION:The increace of Smad3signal and decreasce of Smad7signal may lead to the development of liver fibrosis.The regulation of Smurf1play in the delelopmen of liver fibosis may be limited.Smurf2may play a roleby bidirectional regulation in the progression of liver fibrosis,but the role ofpromoting TGF/Smad singal payhway is outweigh than it’s inhibition.
