Objectives:To observe the effect of electroacupuncture therapy on mitoch-ondrial morphology and function of quadriceps femoris in the IR rats induced by high-fat diet. To explore mechanism that the electro-acupuncture improve the rats with IR quadriceps mitochondrial morphology and function.Methods:48eight-week-old Wistar rats (half-male and half-female), were randomly divided into normal control group (NC,n=16).model control group (MC,n=16),acupuncture treatment group (EC,n=8), sham acupuncture group (SC, n=8). NC given basal feed. MC、EC and SC given high-fat diet for8weeks,then be induced to the model of IR.Using hyperinsulinaemic-euglycaemic clamp evaluated if the models were successful. Then EC received electroacupuncture (EA) on standardized acupuncture pointsthe of Guanyuan(CV4),Zhongwan (CV12),Fenglong(ST30) and Zusanli (ST36). And SC received EA on the non-acupuncture points,each point was inserted in parallel at1-3mm from the Zhongwan acupoint and the Guanyuan acupoint, the Zusanli acupoint or the Fenglong acupoint,respectively. Zhong wan was inserted obliquely with a depth of3-5mm by0.30×13mm stainless steel needle and towarded the pubic symphysis.Guanyuan was inserted obliquely with a depth of3-5mm by0.30×13mm stainless steel needle and towarded the xiphoid, Zusanli and Fenglong were inserted straight with a depth of3-5mm by0.30X13mm stainless steel needle. The positive charge was introduced at the Zhongwan acupoint and the negative charge was connected to the Gwanyuan acupoint,and the positive charge was introduced at the Fenglong acupoint and the negative charge was connected to the Zusanli acupoint. Choosed continuous wave,1mA,2Hz,15min every day,5times every week, for8weeks. The SC received sham acupuncture treatment for8weeks. After the treatment,test the samples.With the transmission electron microscope to observe the quadriceps mitochondria of ultrastruc-ture, shape and number. Using the oxygen electrode to test the respi-ratory control ratio(RCR) and the efficiency of oxidative phosphorylation ratio (OPR), with chemical colorimetry to test the activity of succinic dehydrogenase and the content of adenosine triphosphate(ATP),and the triglyceride(TG) content of quadriceps femoris was measured by enzymatic method.Results:1. After feeding with high-fat-diet for8weeks, then after the treatment,compared with NC.the body weight of MC increased significantly(P<0.05),the TG content of quadriceps femoris increased significantly(P<0.05),the quadriceps mitochondrial morphological structure were damaged for swelling and deformation, the respiratory control ratio significantly decreased(P<0.05), the activity of succinic dehydrogenase was decreased(P<0.05), the ATP content in the quadriceps femoris could also be significantly reduced (P<0.01). 2.After the EA intervention,compared with the MC.the body weight of the EC was significantly lower (P<0.05), the damaged mito-chondria taked restoration and had complete interpenetration of each mitochondrion,the mitochondrial respiratory control ratio improved(P<0.05), the oxidative phosphorylation efficiency is also improved (P<0.05), the activity of succinic dehydrogenase synthase significantly increased (P<0.05), the ATP content in the quadriceps femoris increased (P<0.05), but the TG levels declined (P<0.05).3. After the sham EA intervention, results compared with the MC, no obvious changes in the index of the SC.Conelusions:1.Wistar rats by feeding high-fat diet for8weeks were successfully induced to become insulin resistance models, and the morphology of mitochondria have been damaged varyingly degrees, which can lead to mitochondrial dysfunction.2. The EA treatment can promote the recovery of morphology of the damaged mitochondria, promote the complete interpenetration of each mitochondrion, so enhanced the respiratory function and oxida-tive phosphorylation of mitochondria, increased the utilization of the triglyceride,reduced the weight of the rats,and increased the synthetic rate of adenosine triphosphate.3. The effect of sham EA for quadriceps mitochondrial morphology and function in insulin resistance rats was not obvious.
