| Background and Aims: The pathogenic mechanism and effective treatment ofinflammatory bowel disease (IBD) are still unknown. In the present study, we examined thetherapeutic effects of Chinese Medicine, KUIKELING, and its mechanism on rat colitismodel induced by 2,4,6-trintrobenzene sulfonic acid (TNBS).Methods: Traditional Chinese medicine (TCM) differentiation of symptoms and signs wasanalyzed according to the feature in patients with IBD. Total of 75 female rats was dividedinto normal control group with 12 rats(Group A) and IBD model group with 63 rats. Ratcolitis model of IBD was induced by TNBS. The IBD rats were randomly assigned toGroup B, C and D. Group B was regarded as the model control. Group C was treated withChinese medicine, KUIKELING and Group D was treated with sulfasalazine(SASP) andhydrocortisone (HCT) for 3 weeks. Standard clinical, macroscopic, and microscopic scoreswere used for colitis assessment. Serum cytokine including tumor necrosis factor-α(TNF-α),interleukin-1 beta (IL-1β) were measured by radioimmunoassay and interleukin-10 (IL-10)were measured by ELISA. These cytokines in tissue were measured byimmunohistochemistry.Results: The most patients with ulcerative colitis belonged to the damp-heat type accordingthe TCM theory. Rats that developed acute colitis showed signs of diarrhea, gross colonbleeding and weight loss within 3 weeks. TNBS colitis rats had significantly lower rats ofsurvival than normal control animals. In the control group of TNBS colitis rats, thereshowed that the serum level of TNF-αand IL-1βwas increased. However, the Chinesemedicine, KUIKELING, improved symptoms in TNBS colitis rats. The serum level ofIL-10 in treatment rats with KUIKELING was significantly higher than that of modelcontrol group. In tussue, the expression of TNF-αwas also decreased in the group withKUIKELING treatment. KUIKELING decreased signs of inflammation such as infiltrationby polymorphonuclear leukocytes and multiple erosive lesions.Conclusions: The results suggest that the Chinese medicine, KUIKELING, has potentialtherapeutic utility in experimental rats with inflammatory bowel diseased. The mechanismmay be due to down-regulating the suppressing the secretion of proinflammatory cytokine (TNF-αand IL-1β)and increased the secretion of the anti-inflammatory cytokine (IL-10). |
