Hyperspectral microscopic imaging integrating hyperspectral imaging with microscope is a medical imaging method developed recently. It can obtain images which involve simultaneous pictorial and spectral information on near-distanced biological tissues. And it can provide more plentiful spectral features for analysis than traditional microscope.The paper focuses on the principle and technology of the hyperspectral microscopic imaging. Based on the interferometer imaging spectroscopy, the optical model of the hyperspectral microscopic imaging is studied. In terms of the relationship between object and image, the imaging principle is analyzed. The derivation process and calculation method of the magnification, spatial resolution, depth of field and spectral resolution are described in detail. The algorithm of spectrum reconstruction is analyzed. Around the development of the prototype, the basic structure and working principle of the instrument are discussed. Since the lateral shearing beam splitter is a central component in the prototype, selective analysis is made. The shearing principle is provided and the relationship between shearing value and offset value is given. The derivation process of the size of the lateral shearing beam splitter varying with the incident beam diameter and angle is shown. An experimental prototype is constructed to carry out experiments on the epidermis of broad bean leaves specimen, and the spectral images in visible band are recovered. Experimental results validate the good performance of the system with huge throughput and high spectral resolution, and show that a new method for hyperspectral microscopic imaging is presented.
