Panus giganteus was raw materials in the thesis,the experimental study on the traditional water extraction and ultrasonic extraction method to extract the panus giganteus polysaccharide. The thesis did research in two aspects:polysaccharide bioactivity and chemical structure.In order to further discuss polysaccharide bioactivity and chemical structure interrelation.polysaccharide obtained through separation and purification,and structural analysis.The structural characterization of polysaccharide was confirmed by classical chemical analysis methods with modern analytical techniques and HPLC,GC,MS,NMR,IR techniques. The bioactivity analysis in vitro indicated that polysaccharide possessed certain antioxidant activity and antitumor effect. Establish relationship classical chemical and biological activity.the experimental study provides a new ideas and methods for panus giganteus polysaccharide.The research works are as follows:1.The extraction and purification of panus giganteus polysaccharideThe extraction conditions of hot water immersion:solid-liquid ratio1:30,temperature temperature85℃,time4h,extracting3times.Ultrasonic extraction power:100W,300W,500W,700W;ultrasonic action time20min,extracting3times.Polysaccharide purified by the quaternary ammonium salt precipitation and gel column chromatography separation.2.The antioxidant activity of crude polysaccharideThe chapter mainly detected polysaccharides of antioxidant activity,polysaccharides extracted by hot water and different ultrasonic power. The experimental results show that:five kinds of polysaccharides had antioxidant activity,had strong scavenging DPPH and OH ability,the clearance rate of DPPH reach80%,Ultrasonic extraction of crude polysaccharide clearance rate of hydroxyl free radicals and DPPH significantly increases by about10%than hot water extraction.The total reducing power experimental results show that the polysaccharide had the strong reducing power. Ultrasonic extraction power had impact on polysaccharide bioactivity.Biological activity curve presents parabolic.300W extraction polysaccharide have the highest biological activity.3.The research about acidic polysaccharideWPG1was a kind of acidic polysaccharide,purified by gel column chromatography,can get two components of WPG1-A and WPG1-B,The molecular weight were1.7×106Da、1.6×104Da,Gas chromatographic analysis that two kind of the monosaccharide have similar Monosaccharide composition.two acidic polysaccharides rich in galactose,Mole ratio between monosaccharides are different,By IR of two kinds of polysaccharide have similar molecular bond,Monosaccharide residues by pyran ring form,have-COOH functional group.In atomic force microscopic spectrum,two kinds of polysaccharides form is different,it was found that WPG1-1on mica formed reticulate body aggregates by AFM,while WPG1-2existed in the form of fusiform aggregates structure. The results showed that they were not single strand.4.The research about neutral polysaccharidesSelected the more quality WPG3-A and WPG3-B polysaccharides, was carried on the analysis of the structure,The molecular weight were1.8×106Da4.2×105Da,composed of mannose、glucose and galactose.Mole ratio between monosaccharides were3.3:4.2:7.7and1.05:1.60:5.32.The Carbohydrate chain have two types of glucosidic bonds,they are a glucosidic bond and β glucosidic bond. There are seven'1)Glap(6'and three'1)Manp(4,6'in the backbone,and there are three branch in the main chain, and linked to C4or C6. Image of AFM showed that WPG3-A was single strand polysaccharide, there was no long branch in the main chain.the main chain of polysaccharides of WPG3-B was galactose and glucose constituing by1'6links,have part'1,3,6)links exists in the main chain,mannose is given priority to with the1'and1'6connection,constitutes the branched chain of polysaccharide,'1,3,6) of galactose is the polysaccharide branched chain branching point,Image of AFM showed that polysaccharide WPG3-B with chains and round ball shape form,Therefore WPG3-B of the molecular chain can link can not be sure.5.Ultrasonic effect on structure and activity of WPG3 In this part, selecting the WPG3of300w ultrasonic processing, analyzed the monosaccharide composition and antioxidant activity. Experimental results show that the ultrasonic impact monosaccharides in polysaccharide proportion,at the same time improved the ability of polysaccharide scavenging DPPH.
