Geniposide, one of the main active components from Gardenia jasminoides, can promote bile secretion, regulate gastricfunction, increase the splanchnic blood flow, and it was used as decreasing blood pressure, bacteriostatic, anti-inflammatory, antitumor agent. Previous researches focus on purification of geniposide with macroporous adsorption resin, but there is only few parameter in the large-scale production process. The macroporous resin was commonly used for separation of herb, but the available theory of its absorption is insufficient, it is necessary to find the guide for large-scale production process.Geniposide with the purity higher than90%was obtained in our previous research. SP825-macroporous resin was chosen to further purification of geniposide. The geniposide contents were determined by HPLC. By the use of Response Surface Analysis Methodology, the effects of different factors on the adsorption rate of geniposide were investigated and the optimal intervals were determined. By the method of orthogonal test, the optimum absorption parameters of SP825-resin are determined as solvent concentration at83g/L, pH value at2.79. The final optimum absorption parameters were verified by the dynamic adsorption experiments and ascertained as solvent concentration at83g/L, pH value at3, flow rate at1.5BV/h, ethanol concentration at35:65(v/v), etc.The adsorption equilibrium, thermodynamics and kinetics of geniposide on SP825-resin were investigated under various temperature. The results showed that the absorptive data were well correlate to Freundlich equation. The thermodynamic parameters indicated that the adsorption was an exothermic process, and the dynamic behavior was well described by second-order rate equation, the absorption rate was predominantly limited by liquid membrane diffusion rate. Titration was used to study the surface acid based properties of macroporous resin, the results showed that there were some hydroxylion acceptors on the surface of the processed macroporous resin.In this paper, we obtained the optimum technological parameters of purification with macroporous adsorption resin, and this study has certain prospect of industrial production on the geniposide purification. Also, the adsorption kinetics, thermodynamics of macroporous resin adsorption were obtaind, the basis for further improving the purity of geniposide was provided.
