Organic Catalyzed One-pot Allylation - Ring Synthesis Synthetic High Functional Group Of α- Methylene -γ- Butyrolactone And Of Hydroxytyrosol

The first part of this thesis is the synthesis of highly substituted a-methylene y-butyrolactones via an organocatalytic one-pot allylic alkylation-cyclization (AAC) reaction.Organocatalytic one-pot reaction which is an efficient and environmentally friendly organic reactions strategy is commonly used in organic synthesis. It can maximize reduction the tedious post-processing and avoid the separation of intermediates. Allylation reaction of MBH adducts catalyzed by Lewis base to build a new chemical bond is a frontier approach, and the new compounds are equipped with concentrated and multiple functional groups, a-methylene-y-butyrolactone is a common core structure of many natural products. The allylation reaction between MBH adducts and cyanohydrins obtained allylation products have potential for synthetizing a-methylene-y-butyrolactone. In this part, we designed and developed a new strategy to systhetize the highly functional groups a-methylene-y-butyrolactone. We designed and developed an one-pot AAC reaction for synthetizing a-methylene-y-butyrolactone: it takes cyanohydrins whose hydroxy group are protected as substrates, and then performed with MBH adducts. a-methylene-y-butyrolactone was achieved by the deprotection and cyclization, and the reaction conditions were optimized.The second part of this thesis is to develop a new method for the synthesis of hydroxytyrosol.Hydroxytyrosol is very important fine chemical intermediates, many chemists have devoted themselves to its facile synthesis. We designed a new method to synthetize hydroxytyrosol, which key step is Bouveault-Blanc reaction—the sodium/ethanol reduction of3,4-dihydroxyphenyl acetic acid methyl ester. We successfully achieved the facile preparation of hydroxytyrosol.
