Recently, racetam medicine which can promote brain function, improve the cells to utilize glucose and the ability to study and memory, enhance neuronal metabolism, is becoming a hotspot of the treatment of Alzheimer’s dementia. This paper aims at synthesising Aniracetam, Nefiracetam,and Oxiracetam and studing their polymorphs. We have found that these drugs have better curative effects and less side effects. Therefore, we have the purpose to further prepare their polymorphs, and study their pharmacal properties of the polymorphs. The main contents and conclusions are as follows:In the first part, based on the literature method, we have successfully synthesized Aniracetam, Nefiracetam,and Oxiracetam which also have been demonstrated by elemental analyses, IR, and1HNMR. The improved methods compared with the literature methods are provided lower cost, more environmental protection, and higher security.In the second part, the amorphous states of Aniracetam, Nefiracetam and Oxiracetam are prepared by two methods concluding melt cooling and slow solvent evaporation. The amorphous states have been revealed by IR, HPLC, and DSC. We compared the amorphous states with crystals in stability, solubility, and dissolution rate. The experiment results show that the amorphous states have better solubility and quicker dissolution rate; while the stability of the amorphous states is as much as crystals.
