In recent years, with the development of industry, large amounts of toxic metal ions are discharged into the environment, causing great harm to people’s health. So looking for a method for simple, fast, efficient, sensitive, inexpensive detection environment and organisms of metal ions, neutral molecules is imminent. Fluorescence analysis method, fast efficient, high selectivity, high sensitivity and visual detection based on concept, fluorescent molecular probes are proposed. Molecular fluorescent probes transfer molecular recognition events into fluorescence signals, which can be investigated by analysts. The advantages of molecular fluorescent probes for sensing can be summarized: high sensitivity,"on-off" switch ability, subnanometer spatial resolution and submilisecond temporal resolution, from the impact of electromagnetic field, remote and real time on line sensing, et al, and already have played an essential role in metal cations, anions, neutral molecules, mucleic acids, immune cells and testing.This paper firstly introduces the development history of fluorescence analysis, structure of fluorescent probe molecules, the relationship between fluorescence intensity and structure of fluorescent probe molecules. Secondly, it summarizes the classification of rhodamine and fluorescein probe molecules and their synthesis method. Finally, the literature after the design and synthesis of a series of fluorescein and rhodamine kind of fluorescent probe molecules, and preliminary study on its fluorescent properties.Two fluorescein derivatives were designed and synthesized2-1,2-2. Its structures were a series of characterization. The fluorescent recognition performance was studied, and finding the two compounds have good selective recognition effect for Hg2+range of near neutral or weak alkaline.A rhodamine derivatives3-1was designed and synthesized with rhodamine switch ring structure. The use of rhodamine B ethylenediamine acetamide containing coumarin groups together, make it has the nature of the fluorescent probe molecule itself. When the addition of metal ion, fluorescence resonance energy transfer (FRET) fluorescence probe the mechanism of the probe compounds with select specific recognition effect on Fe3+.
