Maize is one of important cereal crop. Insects attack is an important factor affecting maize production, however, the using of pesticides result in pest resistance has seriously affecting the yield and quality of Maize. The TIFY gene family is found in recent years to enhance plant stress resistance gene which have studied in Arabidopsis, rice, soybeans and other crops. In this study, we used Maize as research materials analyzing corn TIFY gene family by bioinformatics methods. In the following, we select the Arabidopsis thaliana ZmTIFY14gene which is highly homologous with Maize to predicting subcellular localization. The experiments used jasmine acid (JA) and insect bite treatment, and analyzing the relative expression changes. Finally, we provide a gene expression studies on the corn TIFY family to give a scientific-basis on increasing the yield and quality of Maize, the results are as follows:1) Bioinformatics Analysis of Maize TIFY Gene Family:we compared the Maize genome sequence from GenBank and Maize gene sequence website with Arabidopsis TIFY gene family. After the homologous analysis, we found the Maize containing43sets of highly conserved TIF [F/Y] XG amino acid sequence in Maize TIFY gene family. Phylogenetic analysis results show that the Maize TIFY gene family and its homologous genes can be divided into six categories. Maize TIFY gene family has four kinds of genes which have a high degree of homology with the Arabidopsis thaliana by homology comparison. Among them, ZmTIFY14and AtJAZ10have the highest homologous.2The research of Maize TIFY gene expression under JA and inset pest deal. Quantitative PCR technology was taken in this research to detect how many genes of TIFY family express after treated by JA or inset pest. There are two results have been obtained:(1) Under the situation without any outside pressure, there will have no enough JA, so JAZ protein can combined to MYC2, once MYC2had been combined it will lose its function.(2) Under the stress of environment plants can produce enough JA, and these JA can form JA-Ile under the effect of JAR1. Then, JA-Ile can promote the combine of SCFCOⅡ and JAZ, and induce the degradation of JAZ under the effect of26S proteolytic enzyme, the degradation of JAZ can release the dynamic MYC2and start the transcription of JA response gene, so the expression quantity of genes in TIFY family will up at first and will down soon. There are two genes have these features, one of them are named ZmTIFY14, ZmTIFY14is the most sensitive gene to JA and inset pest, its resemble to JAZ10gene of arabidopsis.3The subcellular localization of ZmTIFY14gene:Through bioinformatics analysis and JA and inset pest treat experiment, we found ZmTIFY14gene and AtJAZ10gene are similar not only in its homology but also of their function, both ZmTIFY14gene and AtJAZ10gene can enhance the stress resistance of plants. To fix on their similarity farther, ZmTIFY14gene’s subcellular localization have been done, result show that ZmTIFY14gene localized in cell nucleus just as AtJAZ10gene. So, we can concluded that ZmTIFY14is a important gene related to corn’s stress resistance.
