Cells morphography wasobserved with inverted phase contrast microscope. Cell growth curve of P3andP4passage culture of rabbits BMSCs were drawn according to cell counting.Bone marrow stem cells were stained with blue fluorescent DAPI andfluorescent intensity were observed with immunofluorescent microscopy.Results: The rabbit BMSCs cultured in vitro were adherent and fusiformed,and could formed whirlpool sample structure. The nucleus of BMSCs labeledwith DAPI appeared blue fluorescence and the influence of DAPI on cellproliferation was no significant.Conlusions: BMSCs of high purity could be obtained by the whole bonemarrow adherence culture; the BMSCs cultured by primary cultivation andsubcultivation had good reproductive activity; the BMSCs could be successfullylabeled with DAPI that has no influence on stem cells’ proliferation. Objective: To investige the posibility of establishing choronic radiationinduced lung injury modes in accordance with clinical practice.Methods:12rabbits were randomly divided into two groups:the rightwhole lung irradiation one and the blank control one(normal group). The rabbitsof the irradiation group were irradiated according to the SAD fixed angleirradiation technology. The total dose was40GY and the single dose given wasbased on the radiation dose formula. The animals in the control group werereceived nothing but fed normally. The animals were CT and HE and Massonstains examination at2,4,8weeks after irradiation was over.Results: In the first four weeks, the results of CT were related to the acuteradiation-induced lung in jury. Eight to twelve weeks after irradiation, therewere the ground glass signs, patchy and fiber cord. And the results of pathologysuggested that the thickening of alveolar cell wall and the increase of pulmonaryinterstitial collagen.Conclusion: It is possible to establish the chronic radiation-induced lunginjury models in accord with the clinical practice according to the SAD fixedangle radiation method. Objective: To study the treatment and mechanism of bone marrowmesenchymal stem cell on chronic radiation-induced lung injury.Methods: The rabbits BMSCs were cultured with the whole bone marrowadherent method and then labeled with DAPI. The rabbits’ chronicradiation-induced lung injury modes were established and then divided intoexperimental group and control group. Models in experimental group wereinjected with BMSCs labeling with DAPI by ear marginal vein.Immunofluorescence and immunohistochemistry were used to analyse thetherapeutic effect and mechanism of BMSCs on radiation-induced lung injury.Results: BMSCs were obviously observed in the frozen sections in theright lung of experimental group, but few can be found in the left. HE stainingand PCK examination suggested lung fibrosis condition in the therapeutic groupwere weaker than control group models.
