Purpose: To observe Angelicae Pubescentisand Loranthi Decoction on rats boneinjury and the protective effects of attempts by OPG/RANKL/RANK pathways toexplore its mechanism,through the establishment of type II collagen inducedarthritis SD rats model.Methods:1. The grouping and building:Will only clean level60Wistar rats to forage to ordinary feed a week later,,randomly selected10rats as control group, the remaining50with type II collageninduced model, CIA rats10days later, select the successful rat model, accordingto the random and control were divided into model group, Radix AngelicaePubescentis parasitic Decoction group, leflunomide tablets group, non-steroidalanti-inflammatory drug group and prednisone group,10rats in each group.2. Intervention therapy and materialsModeling success after10days, rats in each group were given Duhuojishengdecoction, leflunomide tablets, loxoprofen sodium tablets and prednisone orally,while the blank group, model group rats were given0.9%saline. All groups werekilled after30days in the administration, taking double hindlimb generalmorphological observation, and shoot bipedal X-ray film, the bilateral ankle,placed paraformaldehyde and coke two ethyl ester in water (DEPC) fixed alternatefixative.3. Observation indicators3.1General observation: Polyarthritis index score standard arthritis indexscore of each group rats.3.2Imaging observation: The foot and ankle X-ray irradiation. Observe andcompare rats foot and ankle in the form of organization change.3.2In situ hybridization detection: Rats ankle bone and surrounding tissueprotection element (OPG), nuclear factor kappa B receptors predominateactivation factor of its ligand (RANKL) mean optical density value. Results:1. Effect of Angelicae Pubescentisand Loranthi Decoction on rat model ofarthritis indexAfter building the model compared with the blank group, the arthritis indexhad significant difference (p<0.01), type II collagen induced the successfulrat model.Compared with the model group, the incidence of Duhuojisheng Decoction groupwere decreased, swelling degree of joint significantly reduced, skin irritation,erythema was not obvious, reduce joint deformation degree. With the model groupin the arthritis index had significant difference (P <0.01).2. Effect of Angelicae Pubescentisand Loranthi Decoction on CIA rat joint X-rayimagingThe difference between model group and blank group: joint space narrowingrelatively obvious visible even disappear, bone osteoporosis,metacarpophalangeal joint fusion. Compared with the model group, DuhuojishengDecoction group rats palm limb joints, interphalangeal joint stenosis degreeis low, bone osteoporosis is not obvious. And X-ray manifestations of leflunomidegroup differences are not very significant.3. Pathological HE stainingAngelicae Pubescentisand Loranthi Decoction group joint cavity were observedwith normal cells in synovial tissue infiltration of inflammatory cells,synovial slightly rough, no necrosis tissue. With leflunomide group,pathological changes basically the same. Angelicae Pubescentisand LoranthiDecoction group and synovial hyperplasia leflunomide group rats is not obvious,the formation of pannus, no cartilage destruction, bone erosion in the synovium,only a small amount of lymphocytes infiltration. Compared with the model group,there is great difference.4. Detection rat ankle the OPG RANKLmRNA expression level.After treatment, compared with model group, the the Angelicae Pubescentisand Loranthi group was significantly higher expression levels of OPG and RANKLexpression levels were significantly lower (P <0.01), OPG and RANKL ratioincreased (P <0.05).Conclusion: Angelicae Pubescentisand Loranthi Decoctionto reduce the CIArats arthritis arthritis index, so as to reduce acute symptoms; reduce CIA ratssynovial tissue inflammatory cell infiltration, reduce pannus formation, thusdelaying the destruction of cartilage, bone erosion; DUHUOJISHENG canelevatedthe expression of OPGmRNA CIA rat ankle tissue to reduce the expression ofRANKLmRNA, thereby improving the ratio of OPG and RANKL, and may delay bonedestruction by this route to reach its role.
