Objective:Electroacupuncture at He sea point-QuChi and Lower he point-ShangJuXu on rat Ulcerative Colitis models, and then observe the effect of electroacupuncture treatment on TNF-α, ChAT, α7nAchR levels, to analyze their mechanism to which they related, briefly explaining the relative specificity of ’He points’ in ’curing viscera diseases by He points’.Method:Forty SD rats were randomly divided into4groups-control groups: blank group, model group, QuChi group and Shangjuxu group. Male and female half a group. The UC rats models were giventrinitrobenzene sulphoneacid(TNBS)/ethanol enema, then "Quchi" group and "Shangjuxu" group respectively received electro—acupuncture at "Quchi" and "Shangjuxu" acupoints, electroacupuncture parameters density wave (10/50Hz), a negative electrode on the right and the positive one left, the intensity between1~3mA, with limb tremor in degrees needle for30minutes once a day for10days.24h after the last treatment,we dissected all rats and collected serum specimen and colon tissue to observe colon ulcer score and the colon tissue changes under optical microscope. Check the expression level of TNF-α and ChAT by ELISA and colorimetric method. Test α7nAchR level by Western blot. Results:1. Naked eye observe the colon ulcer score and observed colontissue structure changes under the light microscope, it shows:①Compared with control group, ulcer score significantly increased(P <0.01) in model group; Compared with model group, the ulcer score of "Quchi" group had a downward trend without significant difference (P>0.05); Compared with model group, the expression of ulcer score significantly reduced(P<0.01) in "Shangjuxu" group; Compared with "Quchi" group, the ulcer score of "Shangjuxu" group decreased significantly(P<0.01).②Under the light microscope, Pathological changes of the Blank group was not obvious;Model group rats colon tissue injury obviously. Obvious necrosis and larger damage, shedding were observed. There are improved changes in and groups and the "Shangjuxu" group is better than "Quchi" group.2. The the serum TNF-α levels, and it shows:Compared with control group, TNF-α significantly increased(P<0.01) in model group; Compared with model group,"Quchi" group made obvious decrease in TNF-α(P<0.01)and the expression of TNF-α in "Shangjuxu" group significantly reduced(P<0.01); Compared with "Quchi" group,the TNF-α lever of "Shangjuxu" group decreased significantly(P<0.01).3. The serum level of ChAT and the colon tissue level of a7nAchR, it shows:①Compared with control group,ChAT significantly decreased in model and "Quchi" group (P<0.01); Compared with model group, the ChAT level of "Quchi" group had a upward trend without significant difference (P>0.05); Compared with "Quchi" group, the ChAT level of "Shangjuxu" group increased significantly(P<0.05).②Compared with control group,a7nAChR significantly decreased in model group and "Quchi" group (P<0.01); Compared with model group,"Quchi" group and "Shangjuxu" group made obvious increase in a7nAChR(P<0.01); Compared with "Quchi" group, the a7nAChR level of "Shangjuxu" group increased significantly(P<0.05). The ChAT is positively correlated with the α7nAchR in every group(r=0.73, p=0.000).Conclusion:①Both of the large intestinal meridian’s lower He point "Shangjuxu" and He sea point "Quchi" can improve the UC through decreasing the ulcer score and TNF-a of the ulcerative colitis and increasing the level of ChAT,a7nAChR.②The treatment effect of "Shangjuxu" on the UC is better than "Quchi",it shows that "Shangjuxu" possess relative specificity.③Our results impartially suggest that "He points" in "curing viscera diseases by He points" mainly implicates the lower He point,and the specific mechanism still needs further research.
