| Objectives:The research is to observe the effect of electroacupuncture on body weight,FBG, GSP,FINS,ISI,Blood lipid(TG,TC,HDL-C,LDL-C) and HPA axis in rats with type2diabetes, in order to prove the interventional effect of electroacupuncture on Yishu(EX-B3) for T2DM in rats, and further explore the potential relationship between the pathogenesis and treatment of T2DM and HPA axis so that it provides experimental evidence for acupuncture treatment of diabetes.Methods:70Male Wistar rats were randomly8rats into blank group,fed with normal diet.The remaining rats were fed with high-fat-diet.After4weeks of dietary manipulation,those rats were treated with intraperioneal injection of STZ at low dose Of35mg/kg body weight.The rats which had FBG and2h OGTT higher than11.1mmol/L were chosen as the diabetic model.If FBG is lower than11.1mmol/L,the rats should continue to feed with high-fat-diet for2weeks.Until FBG dropped to normal levels,the rats were reinjected of STZ at low dose Of35mg/kg body weight for second molding.The successful model rats were randomly divided into model group(containing10rats),glimepiride group(containing10rats),EA on shenshu group (containing11rats),EA on pishu group(containing10rats) and EA on Ex-B3group (containing11rats).Before and after intervention, the caudal vein blood was taken to test blood-sugar content. When the intervention concluded,the rats fasted12h,weighed up and treated with intraperitoneal injection of pentobarbital sodium of30mg/kg body weight. The rats were decapitated quickly to take blood and hypothalamus, pituitary, adrenal tissues were taken out immediately on the ice plate. According to the struction of the kit strictly,the contents of TGã€TCã€HDL-C and LDL-C were measured by colorimetry and the contents of GSPã€FINSã€CRHã€ACTH and CORT were measured by radioimmunoasssay.Results:(1)WT:After4weeks’ intervention,compared with the blank group,the body weight decreased significantly in the model group,glimepiride group and EA group (P<0.01).The body weight decreased in the glimepiride group and EA group after intervention(P<0.05).(2)FBG&GSP:After4weeks’ intervention,compared with the blank group,the levels of FBG increased significantly in the model group(P<0.01).Compared with the model group,the levels of FBG decreased in the glimepiride group(P>0.05),EA on shenshu group(P>0.05),EA on pishu group(P=0.018)and EA on Ex-B3group(P=0.001).The levels of FBG in the glimepiride group and EA on shenshu group were higher than those of EA on Ex-B3group(P<0.05).The levels of FBG decreased in the EA on Ex-B3group after intervention(P<0.05). Compared with the blank group,the levels of GSP increased in the model group (P<0.05).Compared with the model group,the levels of GSP decreased in the glimepiride group and EA group,but there is no significance between intervention and model group(P>0.05).(3)FINS&ISI:Compared with the blank group,the levels of FINS decreased in the model group(P>0.05),while the ISI increased significantly in the model group(P<0.01).Compared with the model group,the levels of FINS decreased in the glimepiride group(P=0.003),EA on shenshu group(P=0.018),EA on pishu group (P=0.005) and EA on Ex-B3group(P>0.05), while the ISI increased in the glimepiride group(P=0.045),EA on shenshu group(P>0.05),EA on pishu group (P=0.002) and EA on Ex-B3group(P=0.001).The ISI in the glimepiride group was lower than that of the EA on Ex-B3group(P<0.05).(4)Blood lipid:Compared with the blank group,the levels of TG decreased in the model group(P>0.05),while TC(P=0.006)ã€HDL-C(P=0.015)ã€LDL-C(P=0.017) increased in the model group.Compared with the model group,the levels of TG decreased in the glimepiride group(P>0.05),EA on pishu group(P=0.004)and EA on Ex-B3group(P=0.001),while the levels of TG in the EA on pishu group and EA on Ex-B3group were lower than those of glimepiride group and EA on shenshu group (P<0.05).The levels of TC decreased in the glimepiride group,EA on pishu group and EA on Ex-B3group,but there is no significance between glimepiride, EA on pishu,EA on Ex-B3and model group(P>0.05),while the levels of TC in the pishu group were lower than those of EA on shenshu group(P<0.05).The levels of HDL-C decreased in the glimepiride group(P=0.009),EA on shenshu group(P>0.05),EA on pishu group (P>0.05)and EA on Ex-B3group(P=0.029), while the levels of HDL-C in the glimepiride group were lower than those of EA on shenshu group(P<0.05).The levels of LDL-C decreased in the glimepiride group(P=0.035),EA on pishu group(P>0.05)and EA on Ex-B3group(P=0.031),while the levels of LDL-C in the EA on Ex-B3group were lower than those of EA on shenshu group(P<0.05).(5)HPA:Compared with the blank group,the levels of CRH(P<0.01). ACTH (P>0.05)ã€CORT (P>0.05) decreased in the model group.Compared with the model group,the levels of CRH increased in the glimepiride group(P>0.05),EA on shenshu group(P=0.016),EA on pishu group(P>0.05)and decreased in the EA on Ex-B3group(P>0.05),while the levels of CRH in the EA on Ex-B3group were lower than those of glimepiride group(P=0.033) and EA on shenshu group(P=0.002).The levels of ACTH increased in the glimepiride group(P>0.05),EA on shenshu group(P>0.05),EA on pishu group (P=0.001)and EA on Ex-B3group(P=0.022). The levels of CORT increased in the glimepiride group(P=0.022), EA on shenshu group (P>0.05) and EA on pishu group (P>0.05) and decreased in the EA on Ex-B3group (P=0.026),while the levels of CORT in the EA on shenshu group were significantly lower than those of glimepiride group(P<0.01) and the levels of CORT in the EA on Ex-B3group were lower than those of glimepiride group(P=0.000)and EA on pishu group (P=0.016).Conclusion:1.EA on pishu and EA on Ex-B3can decrease the levels of FBG,while the effect of EA on Ex-B3is more obvious.It is suggested that back-shu points may have a correlation with nerve segment.EA on Ex-B3can decrease the levels of CORT and excitate glucose sensor in the hypothalamus to decrease the levels of FBG. 2.EA on Ex-B3decrease the levels of CORT so that it can improve insulin resistance,increase insulin sensitivity index and regulate the levels of blood lipid.3.EA on Ex-B3can decrease the levels of CRH and CORT.It is presumed that EA on Ex-B3can act on the hypothalamus and autonomic nervous system subcortical central and motivate senior nerve center integration function so that it regulate the neuroendocrine nerves, neurotransmitter and hormone metabolism.EA on Ex-B3plays an important role in regulating the central nervous system and endocrine function to inhibit the hyperactivity of the HPA axis. |
