Oxidative damage is an important reason for aging body, Alzheimer, Parkinson, cardiovascular disease and cancer and other diseases. Lichen acids and polysaccharide are mainly two categories of chemical composition of lichen, lichen acids with free radical scavenging activity and other aspects of the function, and polysaccharide has excellentent anti-tumor activity. Medicinal lichen originating from Tai Bai mountain are widely applied in Shaanxi folk, including Tai Bai Cha has the efficacy of heat-clearing, detoxification and soothing the nerves; Jin Shua Ba has the effect of anti-inflammatory, pain relief and sedation; Hei Shi Er can play the role of digestive aid and pain relief; Hong Shi Er has the function of digestive aid and inducing diuresis to alleviate edema, four kinds of lichen all have a high development value. However, the research for anti-tumor and antioxidant activity of the above four medicinal lichens is very limited. Therefore, this paper optimized the extraction and purification of the four medicinal lichen polysaccharides, isolated eight kinds of homogeneous polysaccharide, and analyzed these eight kinds of polysaccharides preliminary structural analysis, on this basis, the anti-tumor activity and antioxidant activity in vitro for crude polysaccharides of four lichens and eight kinds of purified polysaccharides were evaluated synthetically, the main content of this thesis was listed as follows:1. A combination of single-factor test and response surface analysis methods, optimizing the extraction method for polysaccharide of Jin Shua Ba, Hei Shi Er, Hong Shi Er, Tai Bai Cha, obtained optimal extraction method of the above four lichens; By comparing three common methods of deproteinization and three kinds of decoloration methods, obtained the best method used to deproteinizing and bleaching processes of the four lichenous polysaccharides.2. Using column chromatography to separate the polysaccharides of Jin Shua Ba, Hei Shi Er, Hong Shi Er, Tai Bai Cha, obtained eight kinds of purified polysaccharides, confirmed both homogeneous polysaccharide; IR scans and the molecular weigh were measured, the monosaccharide composition analysis of eight kinds of purified polysaccharides showed, JSB-1, JSB-2, HEI-1, HEI-2, HSE-1, HSE-2were mainly composed of Man and Glc, while HSE-2contained a small amount of G1cA, TBC-1consisted of Ara, Xyl, Man, Glc, Gal, TBC-2was composed of Man, Glc, Gal.3. JSB, JSB-1, JSB-2, HEI, HEI-1, HEI-2, HSE, HSE-1, HSE-2, TBC, TBC-1, TBC-2had a certain degree of inhibition of proliferation on six tumor cells, which JSB, JSB-1, JSB-2, HEI, HEI-1, HEI-2, HSE, HSE-1exhibited significant antiproliferative activity against HeLa cell, HSE-2, TBC, TBC-1, TBC-2showed significant growth inhibition against Hep G2cell; JSB, JSB-1, JSB-2, HEI, HEI-1, HEI-2, HSE, HSE-1, HSE-2, TBC, TBC-1, TBC-2had preferable antioxidant activity and antioxidant activity, in a certain range, lined in a dose-effect dependent manner.
