Tranexamic acid was used to treat black spots, idiopathic multiple polyps syndrome porphyritic pigmentation and so on, But the way of giving medicine(oral,injection) often cause adverse reactions such as gastrointestinal stimulation.poor compliance,which make it difficult to use; The way is the most direct,convenient and easy to accept for percutaneous treating the skin pigmentation disease, But it’s difficult to break and through the barrier of cuticle and to improve the efficiency of drug percutaneous absorption which make its application is restricted.Considering the pigmentation is mainly local skin lesions, administration may show better prospects. So in this research,we prepared tranexamic acid flexible nano liposome(TA-FNL) gel on the basis of its physicohemical property, and compared the advantages of TA-FNL gel with general gel at the same concentration of ordinary gel at improving the amout of drug penetrated through skin and the skin retention,and studied the promoting permeability of the Chinese medicine natural permeability; Studied the quality of stability, safety on TA-FNL. The aim is to provide theory evidence for the pratical pplication of TA-FNL gel in the medicine and cosmetic medicine field.Set up the analytical method of TA and performed the determination of TA’s solubility and oil/water partition coefficient at25℃.The results showed that the solubility was123.4mg/mL in Phosphate solution (pH6.8), TA has poor lipophilicity, Log P was-678.TA-FNL was prepared by reverse evaporation method, which the optical formulation of TA-FNL suspension was obtained by the central composite design and response surface method, by EE,DL,size, deformation and OD as evaluation index. The suspension such as AV,POV, EE, Size and Zeta were stable at and4℃for2months.To prepared TA-FNL gel and general gel of the same concentration, studying the behavior of penetration though exist mouse skin.The resulst showed thatQ24was1.3times which were significantly more than general gel of the same concentration forTA-FNL gel.In addition,the Qs were almost2.1times more than general gel of the the same concentration for TA-FNL gel.Studied the behavior of penetration though exist mouse skin on the promoting permeability of the Chinese medicine natural permeability; The Q24of TA-FNL has no dependent with the volatile oil of Magnolia,5%volatile oil of Magnolia was significantly improving the Q24and Qs、 so we selected the0.6%TA-FNL gel and to observed the quality standard.Determined the preliminary stability and quality standard on TA-FNL. The preliminary stability showed that the appearance,pH and viscosity,as well as cumulative permeated percentage of TA-FNL gel remained constant.Studied security on the TA-FNL. The results of microbiological test in three TA-FNL gel displayed that the TA-FNL were safe; The results of safety evaluation showed that the TA-FNL gel had no stimulation, sensitization and phototoxicity to the skin.We did’t see the TA-FNL gel in Chinese and Foreign database at the April28,in2013. Form this study,the TA-FNL of prepared can significantly improve the Q24and Qs、though exist mouse skin,solve the bottleneck of transdermal exploitation degree on the conventional transdermal preparations of TA; At the same time,The TA-FNL has good stability, and non-toxic, irritating and allergic to the skin, so this study provides the basis in theory and clinical application for the treatment of skin pigment sink on TA as transdermal preparations.
