The 2H-pyran-2-ones exist in a number of natural products, and achieve broad biological activities, such as anti-microbial property, inhibiting human immunodeficiency virus (HIV) protease or acetylcholinesterase, for the treatment of Alzheimer’s disease and so on. Some naturally occurring compounds possessing 2H-pyran-2-one subunit and many synthesized 2H-pyran-2-ones with simple structures both have anti-cancer biological activities.SK-F12 is a naturally occurring polycyclic alkaloid, which is obtained from marine organism and has promising anti-cancer bioactivity. In the structure, the 5 position of the 2H-pyran-2-one ring is connected to the other part of the molecular. The further study showed that the 2H-pyran-2-one ring is crucial to its bioactivity. Based on the former research work, by means of bioisosterism, 14 target compounds were synthesized and characterized by NMR, MS and IR. Those 14 newly synthesized compounds were not reported in literature.
