Research Purposes:The terminal region of Achilles’ tendon is one of the important parts of humanbody to bear the more stress. The multiple stress stimulations set the calcaneus in thecomplex mechanical environment for a long time, leading that the cell structure changesconstantly and both the long-time excessive stress and unreasonable stress may makethe cell structure change singularly and then necrosis. COX-2、PGE2and their signalingpathways are sensitive to the stress changing. A series of biochemical reactionsproduced by them play a certain role in the development and changing of Tendon-BoneJunction. As the important factor of promoting osteoclast differentiation, RANKLexpression affect the activity of osteoclasts directly. It can be used as the researchcriterion measures for study bone absorption of calcaneus with COX-2、 PGE2.Therefore, this article will research the change regulation of the COX-2、PGE2inTendon-Bone Junction in the condition of jumping, and their relationship with thechange of RANKL in the calcaneus. The article expounds one reason of thepathogenesis of terminal disease from the gene level and provides a gist of the treatmentof terminal disease.Research Methods:This experiment will divide728-week-old male rats into control group (24), jumpgroup (24), loaded jump group (24). They will also be divided into2weeks,4weeksand6weeks group. And then start seven weeks of jumping exercise in the cage withsemi-automatic adjustable voltage. Two weeks after each exercise, killed rats and takenthe blood and the tibia. The area is from middle tibia including the back gastrocnemiusto the middle of feet. HE dyed slice to observe the change of internal tendon bone connection, using the method of immunohistochemistry to determine the number ofpositive cells of COX-2and PGE2in each part of tendon bone connection. And useReal-time PCR to determine the mRNA level of COX-2, PEG2and RANKL in the bonetissue. Analyze the change of OPG, RANKL regulation after stressing.Research Results:1. In HE dyeing section, with the load and time of jumping increasing, jump groupand loaded jump group appeared the typical symptoms of enthesiopathy.2. The differential of the increasing of the numbers of COX-2and PGE2in eachparts of the joint of tendon bone of rats in control group Week2, Week4and Week6has no statistical significance(p>0.05). The expression quantity of COX-2mRNA,PGE2mRNA and RANKL mRNA in calcaneal bone has gradually increased (p<0.05),having statistical significance. The number of COX-2and PGE2positive cells in jumpgroup Week2, Week4and Week6has a significant change (p<0.01) and statisticalsignificance. The expression quantity of COX-2mRNA, PGE2mRNA and RANKLmRNA in calcaneal bone has gradually increased (p<0.05), having statisticalsignificance. The numbers of COX-2and PGE2in each parts of the joint of tendon boneof rats in jump burden group Week2, Week4and Week6has differentials (p<0.05),showing statistical significance. The expression quantity of COX-2mRNA, PGE2mRNA and RANKL mRNA in calcaneal bone has gradually increased (p<0.01), havingdifference and statistical significance. The result of jump group and loaded jump groupare both higher than those of control group. Jump group and loaded jump group have asmaller difference.3.The number of two weeks control group, jump group, loaded jump group of twoweeks rats whose various parts of tendon-bone junction COX-2、positive cells PGE2gradually increasing (P<0.01), with significantly difference, has statisticalsignificance.Rat calcaneus COX-2mRNA、PGE2mRNA、RANKL mRNA expressiongradually increasing (P <0.05), with significantly difference, has statistical significance.The number of four weeks control group, jump group, loaded jump group of two weeksrats whose various parts of Tendon-Bone junction COX-2、 positive cells PGE2gradually increasing (P <0.01), with significantly difference, has statistical significance. Rat calcaneus COX-2mRNA、PGE2mRNA、RANKL mRNA expression continue toincrease (P <0.01), with significantly difference, has statistical significance. Thenumber of six weeks control group, jump group, loaded jump group of two weeks ratswhose various parts of Tendon-Bone junction that positive cells of COX-2and PGE2are gradually increasing(P<0.01), with significantly difference, has statisticalsignificance.Rat calcaneus COX-2mRNA、PGE2mRNA、RANKL mRNA expressiongradually increasing (P<0.05), with significantly difference, has statisticalsignifications.Research Conclusions:1. With the sport time extending, and the increasing of load, the structure in TendonTerminal Area has been changed, appearing some pathologic changes. The reason is thestress make it produce inflammation so that the enthesiopathy happened.2. The pathological mechanism of inflammation which been caused by the stressstimulating, maybe have some contact with COX-2and PGE2factors from theperspective of molecular biology.
