Actinomycetes species is very rich and can produce a lot of active substances used inbiological control and biopharmaceutical, Including agricultural pesticides, antibiotics andanticancer drugs etc. So in recent years puttingt the study of Actinomycetes and its activeproduct has been more widely appreciated. This article mainly conducted a preliminary identifyand study to the Actinomycetes and their antimicrobial activity that were collected from Weishanlake bottom silt.Received a total of21Actinomycetes which collected from Weishan lake after theseparation and purification, carefully observe the colony morphology, spores and spore chain’smorphology and color, combing with physiological and biochemical experiments conducted apreliminary classification and identification. Aroud the Actinomycetes community compositionand their diversity from the three sediment samples zoning of Weishan Lake area, extracted thetotal DNA of Actinomycetes’ single strains and used specific primers to amplify16S rDNA genefragments, built Cloning and sequencing library by digestion and choosed the positive clonescarrying different types of16S rDNA gene insert. Finally, the phylogenetic tree’s build andanalysis combining with the experimental results of the separation, purification andphysiological and biochemical showed: the Actinomycetes that collected from Weishan lake siltsamples mainly distributed in the following four Actinomyces: Streptomyces sp., Nocardiopsissp., Nocardioides sp. and Actinobaculum sp.. Among them, Streptomyces sp. mainly distributedin Nansi Lake District and breeding areas, and then, Nocardiopsis sp., Nocardioides sp. andActinobaculum sp. mainly distributed in the demonstration area.Prepared the21Actinomycetes’ broth and carried out the determination of their antibacterialactivity to different indicator bacteria. The results showed that WS-3and YB-3strains belongingto the same sort appeared the most significant inhibition of the indicator strain, and especially forMildew. At the same time, their inhibition of other indicator bacterias relatively weak. StrainYA-3、YA-6、YC-1、NA-1and NE-2’ inhibition to Mildew second only to the first two strains.Simultaneously, we found that most Actinomyces are able to inhibit the growth ofStaphylococcus aureus and E. coli., but their Inhibition to Bacillus subtilis were generally lower.Initial conditions for the production of WS-3strain’ antibacterial substances were studied, theresults showed that the active substance has a stability to pH, temperature, UV stability, etc. Itcould be determined preliminarily that it does not belong to the protein substances.
