| Microbes in nature always face fluctuant or sudden changes in environmental conditions, such as pH, temperature, nutrients, potential toxic materials, different types of rays, excessive heavy metals, oxides and environmental hormone compounds. For different environmental pressure, fission yeast Schizosaccharomyces pombe can start corresponding genes transcription and expression immediately in response to environmental stimuli for the sake of survive and reproduce. Mitogen-activaled protein kinases (MAPKs) are a type of serine/threonine protein kinase, existing in most eukaryotic cells. From yeast to human the signaling pathway is very conservative, it can respond to a variety of extracellular and intracellular stimulation. It plays a key role in the process of a variety of physiological reactions, including proliferation, differentiation, transformation, inflammation, and apoptosis, etc. In S. pombe there are three MAP Kinase signaling pathways. One of them is pressure activated styl signaling pathway which is activated in a variety of environmental stimulis, such as the osmotic pressure, oxidative stress, heat shock or nitrogen starvation. Many genes are under control of the styl signaling pathway regulation.DJ-1 gene is a kind of new Mitogen-activaled cancer gene, which widely exists in eukaryote. It plays an important role in tumor generation, oxygen free radical scavenging and androgen receptors regulation. In S. pombe, the most related HsDJ-1 homologous protein is SpDJ-1 (SPAC22E12.3c), the other homologs are Hsp3101 (SPCC757.03C), Hsp3102 (SPAC5H10.02C), Hsp3103 (SPBC947.09), Hsp3104 (SPAC11D3.13), Hsp3105 (SPCC1F7.06) and SpDJ-1 has been confirmed to show GLO3 activity. It is indicated that the SpDJ-1 is involved in acetone aldehyde metabolism in fission yeast and playing a role in oxidative stress.On the basis of the predecessors’ work, we use S. pombe as a model to further explore the transcriptional regulatory and expression mechanism of SpDJ-1. First, SpDJ-1 and hsp3105 null strains show significant drawback in survival assay, while hsp3101-hsp3104 null strains entering stationary phase earily. Secondly, we proved that SpDJ-1 is truly regulated by Styl protein instead of Papl. At last, we confirmed the SpDJ-1 gene’s transcription start site and the location of the TATA-BOX. Then we preliminary explored the position of SpDJ-1 promoter’s cis-acting element through bioinformatics and lacZ report gene. |
