In this paper, the main research is the Schizosaccharomyces pombe ribosomal protein L32-1 and L32-2 promoter and transcription factors. Promoters were confirmed by the method of molecular biology and bio informatics. Then building a Schizosaccharomyces pombe cDNA library, find a group of promoter binding protein by yeast one-hybrid structure. The protein can interact with two ribosomal promoters.This laboratory studied RPL32-1 and RPL32-2 different functions in the cell by different methods. The expression level of RPL32-1 and RPL32-2 in cells under different growth conditions are different; When cells enter a state of silence RPL32-1 highly expression inhibition of cell division; While cells in logarithmic phase RPL32-2 high expression to promote cell division.Therefore, previous research found and the functional differences between the homologous genes of RPL32, further proved that the repetitive ribosomal protein homologous genes within the cells have different functions.Ribosomal protein is an important part of the translation mechanism, ribosomal protein gene transcription and translation efficiency directly affects the functions of cells translation mechanism. In this paper, on the basis of previous studies, the promoter and transcription factor was conducted a preliminary exploration. First of all, analysis promoter sequence characteristics of the two ribosomal proteins by the method of molecular biology and bio informatics. The promoter of RPL32-1 is 200bp upstream the transcription initiation site, where, from 120bp to 200 bp exist enhancer sequences. RPL32-2 promoter in the upstream of the transcription initiation point of 1000bp, wherein the presence of 900bp to 1000bp enhancer. Then by the yeast one-hybrid technology, find a group of promoter binding protein RPLX. And determine the RPLX1 and RPL32-2 promoter can interact, and RPLX1 RPLX2 and RPL32-1 promoter can interact with each other.We did not find the ribosomal protein has the effect of promoter binding protein, follow-up experiments to further explore the proteins and binding sites and regulation of the promoter action.
