Hematopoietic stem cells(HSC) capable of self-renewal and the ability to differentiate into precursor cells of blood cells, and generate different cellular blood components, including red blood cells, white blood cells and platelets, they can also differentiate into other cells. Hematopoietic stem cells have two important features that differentiate into all types of blood cells and a high degree of self-renewal or self-replication.Hematopoietic stem cells(HSC) successfully implanted into the bone marrow requires two conditions:(1) HSC capable of homing to the bone marrow hematopoietic microenvironment specific;(2) HSC can be maintained long term in this particular hematopoietic microenvironment. Hematopoietic stem cell survival particular microenvironment called hematopoietic microenvironment, also known as hematopoietic stem cell niche, near the bone marrow hematopoietic stem cells support cells, hematopoietic stem cells involved in the maintenance of self-renewal and differentiation. Bone marrow microenvironment stromal cells, including fibroblasts,macrophages, endothelial cells, reticular cells, fat cells, mesenchymal cells,amplification of these stromal cells in vitro HSC in varying degrees.In order to study the mechanism of HSC and hematopoietic in vitro amplification system, this study:(1) By In Vivo mAb Ly-6C antibody clearance experiments to understand in vivo hematopoietic stem cell expansion mechanism;(2) quantitative detection of mouse hematopoietic reconstruction process bone marrow stromal cells changing trends of gene expression of 22 species provide an intuitive basis for us to identify and hematopoietic stem cell key molecule.(3) Construction of hematopoietic stem cells and stromal cells in serum-free culture system for ex vivo expansion of hematopoietic stem cells provide some theoretical basis and experimental methods.The results showed that mice injected with In Vivo mAb Ly-6C antibody clearance of bone marrow-derived suppressor cells, the main analysis of mouse bone marrow hematopoietic stem cells in vitro, suggesting that bone marrow-derived suppressor cells is oriented cleared after long-term, short-acting hematopoietic stem cells, the expression of pluripotent stem cells(MPP) showed a trend of increase. Indicates mice bone marrow-derived suppressor cells forhematopoietic stem cell proliferation is inhibited in. In normal bone marrow microenvironment, there are positive and negative charge of the regulation of hematopoietic regulatory factors, most of the positive and negative regulator of bone marrow cells by Niche cells, identification within these non-angiogenic factors by quantitative PCR methods expression, can help us to cultivate stem cells in vitro, the use of clear inhibitory factor, add-promoting factor to optimize the culture system.
