PartⅠEXPRESSION OF PDCD4 IN THE ENDOMETRIAL OF MICE EARLY PREGNANCY AND ITS FUNCTIONObjective:The interaction of blastocyst and endometrium is necessary for embryo implantation. PDCD4 involved in many physiological processes including apoptosis, differentiation, DNA damage, conglutination, and it is considered as a tumor suppressor. The results of i TRAQ analysis demonstrated the PDCD4 expression of implantation sites have a significant differences compared with inter-implantation sites, and the expression and function of PDCD4 in mice endometrium during early pregnancy are not reported. In order to elucidate its role in embryo implantation, we designed the following experiment to explore the expression and function.Methods:(1)Explore the m RNA and protein of PDCD4 in mice uteri during early pregnancy and pseudo pregnancy using real-time quantitative PCR, western blot, immunohistochemistry and in situ hybrization.(2)Explore the regulation of PDCD4 in stromal cell decidualization by artificially induced decidualization in vivo/vitro.(3)Explore the protein of PDCD4 in the endometrium of normal female and spontaneous abortion by western blotting and immunohistochemistry.Results:(1)In mice endometrium of early pregnancy, the expression of PDCD4 in D4 and D5 was significantly higher than that in D1, and then it decreased in D6 and D7. Moreover, the expression of PDCD4 in inter-implantation sites was significantly higher than that in implantation sites at D6 and D7.(2)There was no significant difference of PDCD4 expression in mice endometrium during early pseudopregnant in PD1 to PD7. So the expression of PDCD4 was induced by blastocyst.(3)There was no effect on the proliferation and differentiation of stromal cell when silencing the expression of PDCD4. But it is conducive to the decidualization.(4)The expression of PDCD4 protein in female endometrium at progestational stage, decidual membranes and proliferative stage reduce in descending order. The protein expression of PDCD4 in spontaneous abortion endometrium was significantly higher than that in normal decidual membranes.Conclusion:The expression of PDCD4 in mice endometrial of early pregnancy was changed after it was induced by blastocyst. PDCD4 is conducive to the decidualization by promoting the apoptosis of decidualization cell at the right time. Furthermore, the high expression of PDCD4 in endometrium of spontaneous abortion demonstrated the PDCD4 may play an important role in gestation.Part II THE ROLE OF SPOP IN ENDOMETIAL DECIDUALIZATION OF MICEUbiquitination is a regulatory mechanism that occurs after protein translation. To date, few studies have reported on ubiquitination during embryo implantation. In our previous experiments, we used real-time quantitative polymerase chain reaction, immunohistochemistry and western blotting analyses to analyse the expression of SPOP(an adapter of E3 ligases of ubiquitination) in mouse uteri during early pregnancy and pseudopregnancy using an artificially induced decidualization model and a steroid hormone-processing model. The results demonstrated that SPOP may be involved in decidualization. In this study, we established an artificially induced decidualization in vitro model. The results indicated that SPOP influences decidualization by inhibiting over-proliferation of stromal cell. Combined with previous, this study suggests that ubiquitination may be involved in embryonic implantation by influencing stromal cell decidualization.
