With the improvement of people’s living and the increasingly attention to the food safety, natural antimicrobial substance has become the hot topic. Antimicrobial peptide is a kind of natural antibiosis substance, which is generated by biosome. It will cause pathogens to die. And it has been widely used in food, medicine, animal husbandry etc. In this paper the research content mainly include the following respectsThrough the single factor experiment, positive factors were found. And then significant factors were found out by means of PB experiments, including glucose, CaCl2 and ZnCl2. Response surface methodology (RSM) was employed to optimize the cultivation conditions of Brevibacillus laterosporus S62-9 for the enhancement of antimicrobial peptide. A production of 1043 AU/mL was obtained when Brevibacillus laterosporus S62-9 was cultivated at 32℃, pH 7.0, 320 r/min, inoculum size 3%, glucose 15g/L, ZnCl2 8.16 mg/L, CaCl2 1.38 g/L, corn steep liquor 20 g/L on the shake flask level. Futher more, optimization experiments were carried out with the fermenter of 5L. The optimal culturl conditions is cultural temperature 32℃,ventilatory capacity 6 L/min, inoculum size 5%, revolving speed 300 r/min.In the paper, through optimization of culture conditions and culture medium, the fermentation time is cut a lot. And the cost of culture medium is brought down deeply. It will come in to a critical effect to the production practice.
