Xylanase is the enzyme family that dehydrates xylan into xylose oroligoxylose.Xlyanase has extensive applications in the industries of feed, food, fabric and papermaking, and has become one of the hot spots of modern enzymology. This article study on cultivate Aspergillus Niger A09 induction enzyme production using different carbon and nitrogen source induced conditions , we discuss this experiment by Xylanase activity analysis of mycelia and zymogram analysis:xylanase gene F10 and G11 zymogram analysis , relative quantitative analysis, and semi-quantitative analysisthe main content and result of the study were as following:(1) Study on expression rule of the xylanase F10、G11 from Aspergillus niger A09 in different carbon source induced conditions:During the tested carbon source,xylan and xylose is strong inducer on expression of xylanase gene F10 and G11,the induction effect of glucose, maltose, cellobiose is general, Arabia candy and lactose stem two gene expression,relative expression amount of two gene expression of 72 h to the above of 40 h. G11 gene was strong expression in nearly every carbon sources compared With F10 gene expression. In conlusion, when xylan Was the optimal carbon source,G11 gene was strong expression.(2) Study on expression rule of the xylanase F10、G11 from Aspergillus niger A09 in different nitrogen source conditions: During the tested nitrogen source,when Yeast extract、oxalic acid ammonium, NH4NO3 and (NH4)2SO4 were nitrogen source, the expression of xylanase gene F10 and G11was strong, NH4NO3 has the strongest effect, and Peptone, urea, NH4NO3 inhibit two gene expression, F10 gene expressionis below G11 genes in almost any amount of nitrogen source.Thus this experiment xylanase G11 from A09 further optimized medium, choose respectively for optimal carbon source and nitrogen source.Thus we can get the futher optimized medium which was xylan and NH4NO3, Inducing G11 xylanase from the Aspergillus nigerA09 .(3) Study on expression rule of the xylanase F10、G11 from Aspergillus niger A09 in different growth stages: Study on enzyme production rule based on(1) (2) experimental shows that two model xylanase expression rises gradually as time progressed, thus it improve the protein synthesis quantity.xylanase expression amount to the largest level when the Aspergillus niger A09 cultivated 60 hours ,subsequently the two type xylanase expressiongradually decrease as time increases,G11 gene expression is higher than the F10 gene expressionin any time points .Thus we can get the futher optimized cultural time was 60h, inducing G11 xylanase from the Aspergillus nigerA09 .
