Nucleoside derivatives are commonly used for the production and screening of antitumor, antifungal, antiviral and anti metabolic activity, occupying the important role in the life activities. It reports a lot about the nucleoside in the present studies and literatures, effective synthesis of nucleoside derivatives have very important meanings for organic synthesis. The research work of this thesis is divided into three parts.Part Ⅰ:using 2’-deoxynucleosides as raw materials, at first,we protect amino group of nucleoside, react with 4,4’-dimethoxytrityl chloride(DMTrCl) to protect 5’-hydroxyl, then react with 2-cyanoethyl-N,N-diisopropylchlorophosphoramidite to the phosphorus amidation of 3’-hydroxy, forming DNA phosphoramidite monomer, as raw materials of gene synthesis.Part Ⅱ:To study the interaction between PCP protein and upstream A protein, We design and synthesis nucleoside small molecule inhibitors (E)-5’-by-5’-N ({[2-(2-phenyl) ethenyl] sulfonyl} amino) adenosine to simulation "arm" for "small molecule A--PCP" complex, deeply studying their catalytic mechanism.Part Ⅲ:It is a new method for the synthesis of N6-benzoyl -2’-F-2’-deoxyadenosine, firstly using 1,3-dichloro-1,1,3,3-tetramethyldisiloxane to protect 3’-hydroxy and 5’-hydroxyl at the same time, then 2’- hydroxyl fluorination reaction, and the final deprotection of disiloxane, forming the target product.This paper mainly using nucleosides as raw materials, successfully synthesize four kinds of DNA nucleoside phosphoramidite monomer, small molecule inhibitors and fluoro nucleoside derivatives,which were characterized by 1H NMR and ESI-MS data were confirmed.
