Estrogen is a steroid hormone with a similar chemical structure, can directly affect the animal’s metabolism and reproduction。 To achieve rapid and sensitive detection estrogen in milk, microwave-assisted was used to extraction of estrogen in milk and detected by HPLC with UV。1 The optimized conditions of liquid chromatography with UV detected three kinds of estrogenBy HPLC with UV detection device to detect the three estrogen and optimize the three estrogen and optimize the best conditions, UV detection wavelength 230nm。 Solution for HPLC:column temperature 40 ℃, flow rate 1.0 mL/min, mobile phase:A(acetonitrile):B(water)=50:50(V/V), maintain 15min, the sample volume 10μL。2 Extract milk estrogen by microwave assisted extraction technologyBy single factor and orthogonal experiments on microwave extraction and response surface analysis, the optimum microwave extraction is:extraction agent is acetonitrile, solid-liquid ratio of 1:4; microwave time 60s; microwave power 450w.
