Objective:To establish fiber optic sensor instruments applied in drug analysis combided with fluorescence enhancement,synchronous absorption-fluorescence,flow injection and micro sequential injection and lab-on-valve.Method:1.Create a fiber optic sensor fluorescence enhancement device and evaluate fluorescence enhancement effects of fluorescein, rhodamine B, fluorescein, and vitamin B2 and calcein by the condenser mirrors.2.Establish the instrument of synchronous absorption-fluorescence fiber optic sensor and detect the absorption and fluorescence spctrums of rhodamine B, vitamin B2, vitamin B6 compared with UV-visible absorption and fluorescence method.3.Establish the instrument of fiber optic sensor-flow injection in optimal conditions and determine the content of vitamin B2.4.Establish the instrument of fiber optic sensor-micro sequential injection lab-on-valve and measure the content of gatifloxacin capsules by fluorescence method based on the principle of gatifloxacin enhancing fluorescence of congo red-calcein system.5.Establish the instrument of fiber optic sensor-micro sequential injection lab-on-valve and measure the content of cefradine capsules by fluorescence method based on the principle of quenching fluorescein in methanol solvent through optimize experimental conditions.6.Establish the instrument of fiber optic sensor-micro sequential injection lab-on-valve and measure the content of telmisartan tablets by absorption method based on the principle of telmisartan and chloranilic acid charge transfer through optimal conditions.Result:A single condenser mirror enhance fluorescence intensities of rhodamine B, vitamin B2, fluorescein and calcein as a percentage of 151.6%,126.5%, 184.6%,163.8%,respectively;double condenser mirror enhance fluorescence intensities of rhodamine B, vitamin B2, fluorescein and calcein as a percentage of 438.4%,423.8%, 524.2%,552.0%,respectively.2.Synchronous absorption-fluorescence simultaneously measures the maximum absorption wavelength of rhodamine B,vitamin B2 and vitamin B6 at 517.04nm,436.32nm and 293.97nm,respectively;the maximum emission wavelength at 580.13nm,530.93nm and 394.88nm,respectively.Absorption method measures maximum absorption wavelengths at 556.51nm,444.98nm and 289.82nm,respectively.Fluorescence method measures the maximum emission wavelengths at 580.90nm,533.16nm and 393.31nm,respectively.3.Preferred conditions:Flow rate is 1ml/min;reservoir tube length is lm;integration time is 150ms;the average number of times is a second.The maximum emission wavelength of vitamin B2 in 1% acetic acid solvent is at 530.93nm.The fluorescence intensity and concentration within the range of 2.63×10-3-13.17×10-3 mg/ml have a linear equation:AF=1300.2c+417.50,r=0.9997.Recovery is 98.24%-100.35%. Intra-day precision is 1.67%.The tablet made from Guoyao Jituan Xinjiang Pharmaceutical Co.,Ltd.is 54.19 mg/tablet.The tablet made from Shanxi Hengruida Pharmaceutical Co.,Ltd.is 52.27 mg/tablet.The tablet made from Hubei Guangji Pharmaceutical Co.,Ltd.is 54.38 mg/tablet.The fastly measure each sample needing 50 seconds.4.Preferred conditions:The maximum emission wavelength is at 519.73nm.Buffer solution pH is 7.96.The inhaled order is puff gatifloxacin after absorbing calcein-congo red quenching system.The inhalation volume of calcein-congo red quenching system is 10μl.The suction volume of gatifloxacin is 60μl.Fluorescence intensity enhanced by gatifloxacin and concentration within the range of 0.04-0.20 mg/ml have a linear equation:F=2017.5C+255.1,r=0.9992.The precision is 1.04% and the recovery is 96.88%-102.5%.Each an average capsule contains 101.34mg.The accounting to the labeled amount is 101.34%.5.Preferred conditions:Inhalation order is anhydrous methanol,cephradine,fluorescein and anhydrous methanol.The inhalation volume of carrier liquid is 500μl.The inhalation volume of cefradine is 100ll.The fluorescence intensity by quenching fluorescein and concentration within the range of 0.05-0.5 mg/ml have a linear equation:AF=812.6+284.41,r=0.9995.The precision is 4.4925% and the recovery is 101%-103%.Each an average capsule is 258mg.The account to the labeled amount is 103.2%.6.Preferred conditions:Inhalation order is acetone,telmisartan, chloranilic acid,telmisartan and acetone.The ratio of inhalation telmisartan volume before than after is 3:2.The total suction volume of telmisartan is 200μl.The aspiration volume of chloranilic acid is 20μl.The absorbance of telmisartan charge transfer and concentration within the range of 0.064-0.32mg/ml have a linear equation:A=1.505c+0.463,r=0.9996. The precision is 0.3287% and the recovery is 85.2%-105.0%.Each an average tablet is 82.13 mg.The account to the labeled amount is 102.67%.Conclusion:The condenser mirrors of fiber optic sensor fluorescence enhancement device can significantly enhance fluorescence intensity of the fluorescent substance,enhancing the detection sensitivity of drugs.2.Synchronous absorption -fluorescence fiber optic sensor instruments can synchronizly measure emission spectra and absorption spectrum,combining the absorption and emission spectra.3.Fiber optic sensor-FIA instrument through process analysis can quickly and accurately determine the content of vitamin B2,meeting the need of the real-time analysis of industrialization and batch testing.4.Fiber optic sensor-micro sequential injection lab-on-valve instrument through the personalized drug testing and programming can be automated, semi-automated with the traits of online process analysis and reproducible,micro-injection,less agents.
