Due to the large-scale industrial application and huge emissions, CPs(Phenolic compounds) has posed a serious threat to natural environent and human health. This research for 2,4,6- trichlorophenol(2,4,6- TCP) is a typical CPs material as the main object, Set up anaerobic bioreactor system, cultivating domesticated a degradation of2,4,6-TCP capacity of anaerobic granular sludge, examine the change of sludge microbial population structure before and after domestication; In after the success of the domestication, use the intermittent shake flask test and PCR- DGGE(polymerase chain reaction- denaturing gradient gel electrophoresis) technique, examines plus carbon source and metal on anaerobic granular sludge to degrade 2,4,6-TCP efficiency and the influence of the microbial population structure; And further studies the glucose concentration of anaerobic granular sludge degradation 2,4,6-TCP effects and its mechanism.Study the following conclusions are obtained:(1)After 2,4,6-TCP domestication, The PCR- DGGE analysis anaerobic granular sludge microbial diversity decrease, enrichment of six kinds of 2,4,6-TCP degradation bacteria: Thermotog, Petrimonas sp., Escherichia, Bacillus, Clostridium, Burkholderia;Appear new 2,4,6-TCP degrading bacteria: Shigella..(2)Fe added to promote the growth of Escherichia and Clostridium, the two play a crucial role in the process of anaerobic reductive dechlorination. Ni promote Acidobacterium growth, Mn promoted the Mycobacterium growth, at the same time inhibit the growth of Escherichia and Clostridium. This is because the addition of Ni and Mn changed the environment of the pH value of the system, as well as Acidobacterium and Mycobacterium can adapt to extreme environment and degradation of pentachlorophenol, but the degradation ability is poor than Escherichia and Clostridium.(3)Anaerobic degradation process of 2,4,6-TCP is a cometabolism process. Plus the presence of carbon source can effectively improve the anaerobic granular sludge on the degradation of 2,4,6- TCP speed.Promoting effect:Glucose>Sodium Aceate>Sodium Pyruvate>Yeast Extract.(4)The glucose concentration of anaerobic granular sludge degradation2,4,6-TCP has a great influence on the population structure, when the glucose concentration of 2 g/L,2,4,6-TCP degrading bacteria(Enterobucteriaceae bacterium., Escherichia sp., Shigella,Comamonas sp., Clostridium) the dominant position in the microbial system, and2,4,6-TCP maximum degradation rate, but the glucose concentration is too high(>10g/L),using bacteria of glucose, but can’t degrade 2,4,6-TCP(Bacteroides) of a large number of breeding, 2,4,6-TCP degradation rate.(5)In the "anaerobic granular sludge-2 g/L glucose" system, 2,4,6-TCP degradation pathway for: 2,4,6-TCP-2,4-DCP- 4-CP- phenol- mineralization, "anaerobic granular sludge- 10 g/L glucose" system, 2,4,6-TCP degradation paths are: 2,4,6-TCP-2,4-DCP- 4-CP.
