Ginger is a kind of Zingiber Officinale Rosc plant, it contains the volatile essential oil and nonvolatile gingerols. The gingerol which comes from gingerols, is a main functional substance of ginger, and is also the target component of extraction, separation and purification experimental study in this paper.This paper studied the effects of ethanol concentration, temperature, time, solid to liquid ratio and ultrasonic power on ultrasonic extraction of gingerols. And the effects of model of macroporous resin, composition of eluent, ratio of eluent and elution rate on macroporous resin separation of gingerols were studied. And also studied the effects of temperature, absolute pressure, feed rate on molecular distillation purification of gingerol. All of the experimental processes were studied by using Shandong ginger as the raw material. GC-MS, HPLC and H1 NMR were tested, combining optimizes some process parameters with orthogonal test, confirming the optimum operation conditions of extraction, separation and purification experiments respectively. The main experimental results were as follows:1. The extraction experiments of gingerolsThe optimum technological conditions of ultrasonic extraction is as follows: the ethanol concentration is 60%, the extraction temperature is 25℃, the extraction time is 10 min, the solid to liquid ratio is 1:15, the ultrasonic power is 600 w, the extraction yield of gingerols is 5.14%.2. The separation experiments of gingerolsThe optimum technological conditions of macroporous resin separation is as follows: the model of macroporous resin is D4020, the eluent is diethyl ether and n-hexane, the ratio of eluent is 1:1, the elution rate is 1.0ml/min, the yield of gingerols is 82.15%, and the purity of gingerol is 67.04%.3.The experimental study of Molecular distillation purifying the gingerolThe optimum technological conditions of molecular distillation purifying the gingerol is as follows: the distillation temperature is 40℃, the absolute pressure is 200 Pa, the feed rate is 15ml/min, and the purity of gingerol is 86.31%, so the purification effect is very obvious.
