Polysaccharide is one of the main active ingredients of Ginkgo biloba. Ginkgo biloba polysaccharide(GBLP) with anti-virus, anti-radiation, anti-cancer, anti-aging and a series of physiological functions has been recorded in many literatures.Taking yellow Ginkgo biloba leaves as raw material, conditions of extracting and purifing and research of physico-chemical properties of water-soluble polysaccharide from Ginkgo biloba were carried out, the results are as follows:Water extraction and alcohol precipitation polysaccharides were prepared by single factor experiment and orthogonal experiment to determine the optimum conditions. The results were:solid-liquid ratio 1:40, extraction temperature 100℃, extraction time 4 h, extract pH 5.0.Extraction rate of Ginkgo biloba polysaccharide was 12.8% according to the phenol-sulfuric acid method.Protein was removed by sevag method. Polysaccharides from Ginkgo biloba leaves were isolated and purified by DEAE-52 cellulose column chromatography, two acidic polysaccharides were obtained after elution with 0.2 mol/L and 0.3 mol/L NaCl solution, The main one was purified by gel exclusion chromatography Sephadex G-100 further.GBLP molecular structure and molecular weight were analyzed by Fourier Transform Infrared Scanning(FTIR) and matrix-assisted laser desorption time of flight mass spectrometry(MALDI-TOF-MS) respectively. The analysis results indicated that there were multiple characteristic absorption peaks in FTIR. The molecular weight of GBLP was 13 kDa through MALDI-TOF-MS analysis.Vc as the positive control, GBLP vitro antioxidant activities including scavenge hydroxyl radicals, superoxide anion radical, DPPH radical ability were studied. IC50 of them was 4.91mg/mL, 1.77 mg/mL, 0.36 mg/mL respectively. This suggested that GBLP is a new type of natural potential antioxidant.Glycosylation of soybean peptide complex is synthesized from GBLP and soy peptides through Maillard reaction, which have higher antioxidant activities than the controls.
