Micromonospora carbonacea JXNU-1 is a strain of Actinomycete with board-spectrum antimicrobial activity, isolated from a soil sample of farmland by our lab. M. carbonacea JXNU-1 can produce a single-component antibiotic which has a importantly broad development prospect. The fermentation process in the 5L fermenter, the physicochemical property and adorption characteristics of bioactive products of M. carbonacea JXNU-1 was studied in this paper. The main results were as follows:(1) The fermentation process of M. carbonacea JXNU-1 in the 5L fermenter was optimized by single factor experiment test. The results showed that the optimum fermentation conditions were at temperature of 30℃, initial pH of 7.7, ventilation of4L/min, stirring speed of 250 r/min, 6% of inoculation proportion and 2.5L of liquid medium in a 5L of fermenter 96 h of fermentation;(2) The effect of concentration, hours, quantity and times of refilling glucose on the fermentation production of antibiotics from M. carbonacea JXNU-1 was studied by single factor experiment test. The results showed that 3.5g/100 ml of glucose concentration was fed one times at 28-36 h, the quantity is 300 ml, the fermentation productivity increase obviously.(3) The stability of antibiotics from M. carbonacea JXNU-1 was studied, and the results showed that the antibiotics was stable at high temperature,alkali and light,but sensitive to acid.(4) The effect of the iron exchange resin, the time of adsorption, the initial PH,and the concentration on the adsorption of antibiotics from M. carbonacea JXNU-1was studied by single factor experiment test. The results revealed that the 717 anion resin was the optimal resin for the adsorption of antibiotics from M. carbonacea JXNU-1, and the adsorbance was the best at the acidic range of pH(pH 5) with the adsorption time of 2.5 h. The Freundlich model was fit well.The results mentioned above laid the solid foundation for further research on antibiotics from M. carbonacea JXNU-1.
