With the continuous improvement of people’s living standard and the change of the diet, the incidence of hyperlipidemia increases year by year.Dyslipidemia is a metabolic disorder, total cholesterol (TC), triglyceride (TG), low density lipoprotein cholesterol (LDL-C) and high-density lipoprotein cholesterol (HDL-C) one or more of an exception occurs, it is a major risk factor for cardiovascular morbidity and mortality.According to statistics, by controlling the serum total cholesterol (TC) to interfere with the sclerosis of arterial congee appearance, which can reduce the cardiovascular worse.Pomegranate ellagic acid is important bioactive components of pomegranate, have fall hematic fat, antioxidation, anti-cancer, antibacterial and so on many kinds of biological function, this article to health golden hamster hyperlipidemia animal model for research object, with pomegranate ellagic acid as subjects, research on the golden rats blood lipid and liver cholesterol synthesis and HMG-CoAR clear related gene mRNA, SREBP-2 mRNA, LDL-R mRNA expression, the effect of pomegranate ellagic acid on experimental hyperlipidemia golden gophers ability to influence and cholesterol synthesis and clear.This experiment adopts the high fat feed on normal hamsters fed continuously 4 weks to induce high-fat animal models, then divided into normal group, model group, pomegranate ellagic acid high, medium and low dose group and positive control group, in addition to the normal control group and model group, the rest of the group continuous lavage corresponding subjects 4 weeks, observe its effect on blood lipid level, at the same time, the rate of ultraviolet spectrometry method is used to detect liver HMG-CoA reductase activity, real-time PCR method (RT-PCR) detection of liver cholesterol synthesis related genes and low density lipoprotein receptor related gene expression level.The results of the study are as follows:(1) This test choose golden mice as experimental animals, by feeding high fat feedstuff containing 15% lard,0.1% cholesterol to build mold,4 weeks after feeding high fat forage, successfully established the golden experimental hyperlipidemia Hamster model.(2) The pomegranate ellagic acid can significantly reduce the experimental hyperlipidemia in rat serum total cholesterol (TC), golden total triglyceride (TG) and low density lipoprotein (LDL-C) level, the marked increase in high-density lipoprotein (HDL-C)levels, prove that pomegranate ellagic acid has significantly cholesterol-lowering effect.(3) Based on experimental hyperlipidemia in golden mouse liver HMG-CoA reductase activity and HMG-CoA reductase mRNA expression of impact analysis found that pomegranate ellagic acid may be the mechanism of reducing cholesterol synthesis by inhibiting HMG-CoA reductase activity, instead of dampening the HMG-CoA reductase mRNA expression.(4) The hamster liver on experimental hyperlipidemia golden land of LDLR mRNA and the effects of SREBP-2 mRNA gene expression analysis found that pomegranate ellagic acid lipid-lowering effect of the other way is through the SREBP-2 mRNA, which raised its downstream gene mRNA expression of LDLR, thus increasing the liver cell surface of LDLR, strengthen the liver cells in peripheral blood of LDL-C endocytosis removal ability and realize the lipid-lowering effect.In a word, Pomegranate ellagic acid has significantly reduced experimental hyperlipidemia animal blood lipid levels, especially can significantly reduce blood total cholesterol and triglyceride level, at the same time, can significantly increase the liver cells, low density lipoprotein receptor gene expression, thus improve the swallowing clear action within the liver of low density lipoprotein cholesterol, its mechanism may be mainly through raising SREBP-2 mRNA expression, thus raising its downstream target genes of LDL-R mRNA expression.
