Mycoporine-like amino acid is a kind of specific compound which exists extensively in marine algae organisms. It is a naturally high-efficient ultra-violet absorber. Because of the depletion of the ozone layer, seeking a naturally high-efficient ultra-violet absorber to replace the artificially synthesized products is becoming a hotspot currently. China is rich in marine resources, the annual output of the algae is millions of tons. Therefore, exploring a new technique to extract mass mycosporine-like amino acid from algae is of great significant.In this paper with Qingdao porphyra yezoensis as material, (1) Optimized the extraction process of the MAAs compounds by single factor and orthogonal experiment; (2) improved the purity of MAAs compounds in the samples by resin separation experiments; (3) established a technique to extract high purity MAAs compounds efficiently.1. In this paper, the major MAAs ingredients from Porphyra yezoensis were analyzed and identified using the UV spectrum scanning techniques, liquid chromatograph, liquid chromatograph-mass spectrometry. the results showed that two kinds of MAAs were identified, namely:Shinorine and Porphyra-334. Their molecular weights were 332 and 346 respectively.2. By single factor and orthogonal experiments, the optimum extraction process is determined as:ethanol concentration is 15%, extraction temperature is 35℃, extraction time is 2 hours and the ratio of liquid and material is 40(g/mL). Under optimal method, the extraction ratio of porphyra-334 is 14.7mg/g.3. The purification process of MAAs compounds by 001×7 resin includes the following steps:pH value of the sample is 6.0, concentration is 7mg/mL,the sample flow ratio is 2BV/h(column volume/h), the sample volume is 5BV. The eluent is PBS contains 10% NaCl, the elute ratio is 3BV/h(column volume/h). Liquid was collected under vacuum freeze drying.
