Phenylglycine as a kind of important drug intermediates, it has optical activity. D-phenylglycine as a drug intermediates, commonly used in ammonia penicillin’s preparation such as antibiotics.At the same time, L-phenylglycine as resolution agent often used in many organic reactions, it is also as a important intermediate for the synthesis of some pesticides. Due to its widely range of applications, the pharmaceutical products can be made into a variety of dosage forms for the body to absorb, it has a broad prospects in the field of medicine. How to continuously improve the quality of phenylglycine products, reduce the production cost is particularly important. And crystallization process is one of the important factors affecting product quality, therefore, this article mainly research in optimization of the phenylglycine crystallization process.In this paper, the phenylglycine crystal structure was studied, phenylglycine was analysed by infrared spectra and X-ray powder diffraction, use the Materials studio software to analysis phenylglycine, obtained the phenylglycine crystal cell parameters, crystal system and space group, determine the crystal structure of phenylglycine. By BFDH and AE model successfully predicted the phenylglycine crystal habit.The development and design of crystallization process cannot be separated from its related thermodynamic properties, such as solubility, metastable zone, the quality of the product are also affected by thermodynamic properties. Therefore, we need to determine phenylglycine solubility in the solution and the metastable zone.In this paper, a laser dynamic method is use for the determination of phenylglycine solubility in pure water, methanol-water, ethanol-water, different pH water, also uses this method for the determination of the metastable zone.Then, uses the Modified Apelblat equation associates the phenylglycine solubility in water points, the R-K equation associates the phenylglycine solubility in the methanol-water and ethanol-water mixed solvents. At the same time, phenylglycine were measured under different temperature of metastable zone, which provides the theoretical foundation for the development of crystallization process.Finally, according to the above study of crystal structure, crystallization thermodynamics, with comprehensive consideration, we choose the best crystallization process. By investigating the solution analysis agent content, crystallization temperature, crystallization of pH, the rate of flow speed and raise the conditions such as crystallization time influence on the purity and yield of the phenylglycine product. Determines the optimum process parameters of crystallization.
