Soluble sugar has been found to be not only one of the important nutrients in cereals and legumes, as well as extremely beneficial physiological effects on the human body. Although soluble sugar in cereals, legumes were low levels, which in plant growth, characterization, processing production and genetic breeding played an important role. However, there is no uniform method for determination of soluble sugar in cereals, legumes. Even if the same way, but with different choices of reagents, preparation methods, environmental conditions, and the determination of the parameters, the difference between the results may be also large. Therefore, an urgent need normative method proposed to improve the accuracy of measurement results.This paper is intended to research on determination of soluble sugar in cereals and legumes by shaffer-somogyi. Some representative grains, legumes were choosing as samples for this study, smashing degree of samples, preparation of reagents, optimization the measured parameters and analysis of several conditions, etc. were conducted. By measuring the standard curve, accuracy, precision, and other indicators of this method to establish a more accurate and convenient for determination of soluble sugar in cereals and legumesThe conclusions were as follows:Measurement methods and standards of detecting sugar was referenced, after optimized and validated this method, the determination method of soluble sugar in cereals and legumes was given. The results showed that it was accurate and feasible.Single factor experiments were conducted optimized the samples preparation and processing parameters including some critical conditions. Optimized results are: extract of soluble sugar was 80% ethanol, 80 ℃ water bath for 30 min as extraction conditions; total soluble sugar acid hydrolysis conditions for hydrolysis 80 ℃ water bath for 30 min; protein precipitating agent for zinc sulfate – potassium ferricyanide; crushed and sieved sample pretreatment selection of 40 mesh; sample handling degreasing step can be omitted.After through six collaborating laboratories verified this method. The linear range of the standard curve method was 0 mg ~ 2.5 mg, recoveries rate were 95.64 % ~ 101.17 %, under repeatability conditions precision was 0.65 % ~ 3.76 %, the precision under reproducibility conditions was 0.99 % ~ 3.53 %, repeatability standard deviation was 0.053 %, reproducibility standard deviation was 0.076 %. This method of shaffer-somogyi used to determine soluble sugar in cereals and legumes revealed that repeatability standard deviation, standard deviation absolute difference reproducibility, repeatability limit, reproducibility limit. Among these test results were not greater than the repeatability limit and reproducibility limit, it was acceptable, and this indicated that the method is more accurate.This paper filled the blank about determination of soluble sugar in cereals and legumes in China. This method is simple, easy to operate; also the efficiency of the analysis can effectively improve. It provides research evidence that determination soluble sugar content of cereals and legumes for national standards.
