Propolis, a kind of natural products, was used to fight against pathogenic microorganism invasion. Propolis has a lot of biological compounds and a variety of pharmacological properties. The antioxidant property is one of its most important pharmacological properties. It’s great significance that research on activity ingredient identification and antioxidant property of propolis. Therefore, in this paper, including components analysis and antioxidant property of propolis from China was studied. A variety of antioxidant methods and cell model of in vitro antioxidant was used to conduct a comprehensive evaluation. Studying the relationship between the antioxidant property and components of propolis, and compared the differences of different methods.Using raw propolis from 85 areas of China as material, total phenolics and total flavonoids content in extracts of propolis by 75% ethanol-water were estimated and the preliminary qualitative and quantitative analysis of components in extracts of various regions was done by using HPLC, and the propolis was classificated based on composition and content of propolis. The contents of total polyphenols and flavonoids of propolis collected from different areas varies, Hunan province has highest total phenolics and total flavonoids content in extracts, with 319.77mg/g raw propolis. By identifying the components in extracts, we found that 6 kinds of flavonoids, which were pinobanksin-3-O-acetate, chrysin, pinocembrin, pinobanksin,galangin,5-methoxy pinobanksin,with high content in extracts of mostly raw propolis. Caffeic acid phenethyl ester, P-coumaric acid, caffeic acid and isoferulic acid have higher content in extracts of mostly raw propolis compared with other kinds of phenolic acids and their esters. Propolis from the Chinese mainland was divided into five categories based on HPLC chromatography figure features, most of them belonged to previous three categories. Class A has highest total phenolics and total flavonoids content in extracts, with 184.74mg/g raw propolis.Propolis from the Chinese mainland extracted with water,75% ethanol-water, petroleum ether, ethyl ether, ethyl acetate. The comprehensive antioxidant capacities of the hydrophilic and lipophilic extracts of propolis were evaluated by ORAC assay and anion radical scavenging activity. The antioxidant properties of propolis extracted with different extraction solvent listed from strong to weak as follows:75% ethanol-water extract>diethyl ether extract, ethyl acetate extract>water, petroleum ether extract. And the antioxidant properties of five categories listed from strong to weak as follows:A>B, C, D>E. The antioxidant capacities of propolis extracts showed strong positive correlation with the total polyphenol content. The antioxidant capacity of propolis is enhanced with increasing polyphenol content.The antioxidant activities of different extracts of propolis were investigated by cell-based antioxidant methods used flow cytometry, fluorescence microplate reader and fluorescence inverted microscope. The results showed that all extracts of propolis had strong antioxidant activities. And propolis extract in Heilongjiang baoqing had the strongest cellular antioxidant activity.
