| In contemporary society, various kinds of false advertising of fake and shoddy products appear on the market because of the rapid development of health food. So the related department appear a hole in the supervision and management of health care products. At the same time, the safety of Health Foods has become the focus of attention of the social and government, and also cause the concerns of consumers.Therefore, it is essential to strengthen the understanding of health food,and solve the problem of prohibited drugs added into health foods correctly, especially establish a perfect testing method of illegal additives in health food. Traditional analyst usually use the mothed of high performance liquid chromatography(HPLC) or high performance liquid chromatogaphy- mass spectrometry(HPLC-MS) to detect illicit drugs for qualitation and quantification in health foods. The advantages are high sensitivity, and accurately. However, these methods are time consuming and need sample pretreatment intricately. In addition, the mobile phase needs a large number of organic reagents. As a result, no matter from the perspective of economic and environmental protection, they are not practical in the actual detection of large quantities of samples.The development of science and technology has driven the development of mass spectrometry. As the hotspot and frontiers among mass spectrometry technology, ambient ionization technique has a fast analysis speed without complex test samples pretreatment. What’s more,it can be directly ionized for amounts of complex matrix samples, greatlyimprove the efficiency of mass spectrometry. Paper spray ionization is a new branch of ambient ionization mass spectrometry, it has excellent development prospect and broad application field with outstanding superiority such as easy to operate, low cost and less solvent consumption.In recent years, the paper spray ionization technique has been widely used in environment, food, drug administration, public security and other fields of research at home and aboard. The main content of this paper is as follows:(1) Rapid determination of three PDE5 inhibitors including sildenafil, vardenafil and tadalafil in the anti-fatigue health food by paper spray ionization mass spectrometry. In positive ion mode, under the voltage of 3.5 kV, the ion source temperature of 100 ℃, with methanol as solvent, the standard substance is tested first, then practical samples are tested for qualitative analysis with 20 μL each time. Finally, quantitative analysis is typically achieved by the signal intensity ratio of measuring material and internal standard substance sibutramine. The results show that the method of testing the three PDE5 inhibitors added in the health food as semi-quantitative analysis results believable. It showed good linear coefficient in the range of 5.00~56.00 mg·L-1 for the three target analyte. The relative standard deviation is higher than 0.99. The limit of detection is lower than 0.1 mg·L-1. Compared with HPLC quantitative results, the semi-quantitative results of this method really exist certain gap, but as a result of semi-quantitative is workable, and the pretreatment is simple, analysis quickly and without time-consuming.(2) Under the optimized conditions, the high-throughput screening of illegal additives in weight reducing health food through sprayionization technology is able to realize. Five kinds of common illegal additives including sibutramine, single methyl sibutramine, double to methyl sibutramine, phenolphthalein and fenfluramine are checked out in this method successfully. In positive ion mode, the quantitative analysis is carried out by measuring the signal strength of the determinand and the internal standard-sildenafil. The results showed that the method of five kinds of banned substances in the market losing weight health food as a semi-quantitative analysis believable.(3) Developed a new analysis method of mass spectrometry, and compared the results of hydrophobic paper and plain paper in negative voltage mode. Finally, the hydrophobic paper was applied to analyze the banned additives in fall blood sugar health products for rapid screening as well as semi-quantitative detection. Six kinds of sulfonylureas material including glibenclamide, gliclazide, glimepiride, gliquidone, glipizide and pioglitazone in different dosage forms including tablet, pill and capsule are tested respectively. |
