In this paper, the citrus peel as raw material, the combined extraction process, separation, purification and determination methods of pectin and hesperidia were studied.Compared the advantages and disadvantages of alkali extraction process and the alcohol extraction process. Eventually determine choose alcohol extraction process to extract pectin and hesperidia in citrus peel.Some factors such as Methanol concentration, extraction time, liquid crystal pH, pH value on the yield of hesperidia were investigated. The optimum process conditions are obtained through the test, which are the mass fraction of 99.5% methanol, times 2.5 h, the crystallization solution pH of 4,the sedimentary solution pH of 4.5. At this time, hesperidia extraction yield is 6.16%.after washing with ethanol, and finally get lighter color, higher purity of pectin products. the research shows that: “methanol method ~acid method” process obtain the most optimal products quality of pectin and hesperidin, next is “acid method~alkaline method” process, and the worst is “alkaline method~ EDTA method” process. The study of “methanol method~acid method” process found the optimal condition is that: methanol concentration of hesperidin extraction was 99.5%, the pH of crystallization of liquid was 4, primary crystallization time was 2 hours, hesperidin extraction time was 2 hours, rinse material liquid ratio is 1:7, rinse for 30 min, bleach concentration was 0.1%, the extraction liquid ratio of 1:10, pH of extract solution was 2 hours, extraction temperature was 90℃, pectin extraction for 2 hours, and the yield of hespridin and pectin were 2.79% and 12.58% under given condition,respectively.
