In recent years,there was a large number of relevant reports about rapeseed water enzymatic oil extraction technology in domestic and foreign,but because of the variety of enzyme and add quantity, hydrolysis reaction is a long process, emulsification degree factors such as serious and reduce the oil extraction efficiency, which seriously restrict the water enzymatic oil extraction technology industrialization promotion. The enzymatic preparation process water rapeseed oil were studied optimized the broken wall enzyme and protease hydrolysis process, and studied the emulsion breaking methods, improve the oil efficiency. Then this paper studied the enzymatic extraction of rapeseed oil nutritional and functional activity evaluation, explore the hydrolysis process on the quality of oil, and the physical and chemical indicators and fatty acid composition differences in enzymatic extraction rapeseed oil between the other methods.Research mainly includes four aspects:Water enzymatic broken wall process;Protease hydrolysis process optimization; Emulsion demulsification process; Rapeseed oil supplements qualitative evaluation.Water enzymatic broken wall technology research include preparating enzyme and optimizing hydrolysis process conditions,the results showed that Pectase NCB-PE40 was the best at broken wall effect, by means of the response surface method was optimized,the results showed add enzyme content was 1.00%, enzyme solution temperature 50℃, the proportion between liquid and material was 6:1, hydrolysis time was 3 h, hydrolysis solution pH4.5 conditions, the rapeseed oil efficiency reached 95.13%.In the hydrolysis process of broken wall, on the basis of optimization protease hydrolysis process, get rapeseed oil. Through the alkaline protease Alcalase 2.4 L, alkaline protease Protex 6 L and neutral protease Protex 7 L screening, found that the alkaline protease Alcalase 2.4 L enzyme was best, oil yield was the highest. Through the single factor and response surface test, optimize the protease hydrolysis process, the results showed add enzyme quantity was 1.50%, enzyme solution temperature was 60℃, hydrolysis time was 100 min, enzyme solution pH9.0 conditions, oil yield reached 62.31%.After enzyme solution, some macromolecular material such as fat, proteinetc from rape seed cells to the water phase. Due to their polarity, viscosity and hydrophilic close lipoprotein (surfactant) and allows for the existence of shouldn’t miscibility of oil and water together, is not easy to separating completely. In order to improve the oil yield, ensure the quality of rapeseed oil, it is necessary to emulsion for emulsion breaking. Through the demulsification research showed that, centrifugal, quiet place, freezing and thawing were helpful in emulsion breaking. Optimization of centrifugal emulsion breaking condition, get the best demulsification process for. static buy frozen 36 h with the temperture 50℃ and thencentrifugal time was 25 min, centrifugal speed at 8000 r/min, centrifugal pH 3.5, the emulsion of emulsion breaking rate could reach 46.5%.In order to further evaluate oil nutritional value, verify the enzymatic extraction water the feasibility of rapeseed oil process, this study compared the enzyme solution and enzyme solution without the rapeseed oil and fatty acid composition of various physical and chemical indicators.Found that in addition to acid value, peroxide value and phospholipid three indicators before and after enzymolysis slightly fluctuation outside, the rest indexes have no obvious changed, before and after hydrolysis oil fatty acid composition and content that is relatively stable, indicating that water enzymatic process feasibility. Experiments and compared with water enzymatic, line type extraction and press method for rapeseed oil physical and chemical properties and the influence of the fatty acid composition, and the results showed that three methods from the oil in the acid value, peroxide value and phospholipid content have obvious difference outside, the rest indexes was not significantly different.
