Magnetic microspheres as a new type of composite material, they have some advantages of the surface design, targeting, repeatable recycling. In recent years, is widely used in academic research.Through a combination of magnetic microspheres and glutaraldehyde, they can connect to the targeted drugs, significantly so that it functions to play.This project is based on corn starch as starting material, occurring in role of amylase by enzymatic hydrolysis.thus porous starch microsphere.The experimen.by optimizing conditions of enzymatic hydrolysis, we can know the best conditions for porous starch formation is found at temperatures of 50℃, pH of 3.5, the amount of added enzymes is lml/20g, the enzymatic hydrolysis time in about 24 hours. Fe2+ Fe3+ mixes under weak bases, coprecipitation reaction system with magnetic particles of Fe3O4. Fe3O4 is mixed with porous starch using centrifugal shocks several times, to be injected into the pores of porous starch microspheres, allowing the starch microspheres have magnetic effect, after washing several times, resulting in the necessary magnetic microspheres.Through covalent method, magnetic porous starch microspheres as a carrier, ALDC fixed on the carrier surface, it can receive the immobilized ALDC. Further analysis of the results showed those:when amount of enzyme addition is lOmL/g, when amount of glutaraldehyde as a total volume of 1%, the enzyme immobilization is best. In this way, the protein immobilization of ALDC is 65.59 mg · g-1, specific activity is 745.39 U/mg-1, recovery rate of 40.85%. In addition, the ALDC immobilization at 25℃, pH 6.0 environment, the highest activity of the enzyme, and has a thermostability and pH stability.This issue is not limited to this feature of a-Acetolactate decarboxylase enzyme research of single, but proved in experiments of magnetic microsphere in the contemporary value embodied in the hard sciences.Believe that,with the advancement of technology,magnetic microspheres will play its unique role in various fields,it will also provides a theoretical basis on commercial industrial production in the future.
