With the continuous progress of the era, the attention of health food is gradually being made by the majority of the people. But in recent years,some unscrupulous producers want to achieve the purpose of obtaining huge profits, they are illegal to add variety of ingredients in health food.The results lead to a decline in quality of health products, and even additives itself may produce toxic effects of carcinogenic and mutagenic.Because illegal add has strong concealment, its detection has become a challenge task to maintain health food safety. In this paper, the extract of Ginkgo biloba leaves illegally added rutin,and illegal add synthetic industry pigment in green tea, has launched the analysis method of the research, the specific contents are as follows:1. A LC-MS/MS qualitative and quantitative method for determination of rutin and quercetin content in Ginkgo biloba leavesBy convection dynamic phase composition and extraction solution conditions, we optimized the best UHPLC-MS/MS determination conditions,0.1% acetic acid solution and acetonitrile as mobile phase,using a gradient elution program, ESI source,using negative ion scan and multiple reaction monitoring(MRM). This method was used to analyze the 19 batches of ginkgo leaf samples from different producing areas.2. Near infrared spectroscopy method for calibration and prediction of the rutin content and the mathematical model is established.Collecting the NIR-spectra of 19 ginkgo biloba leave samples from different origin areas,multiplication signal correction and first derivative were conducted to optimize the original spectra with OPUS 6.5 software.In7500-6099 cm-1and 5450-4249 cm-1 range inside, the rutin content was quantitative detected quckly with Partial least squares( PLS)by choosing seven principal components, a rutin quantitative mathematical modelof ginkgo biloba leaf based on ultra performance liquid chromatography-tandem mass spectrometry results was establish. The near infrared model prediction results match to the chromatographicdetection results, model variance were 0.194 and the average recovery of predict ion samples was 98.40%. This method is rapid and simple, it can be used to quality control of Ginkgo biloba medicinal and high-volume products detection.3. Determination of organic dyes in teaA method was developed to qualitatively and quantitatively analysize the illegally added of Fast Green, Lissamine Green, Patent Blue and Brilliant Blue in green tea by using ultra performance liquid chromatography- tandem mass spectrometry. Due to the complexity of the tea matrix, we optimized the best UHPLC-MS/MS determination conditions and solid phase extraction conditions. Through the establishment of the dye method which may be added into the tea, it provides a more comprehensive method for the detection of organic dyes in tea.
