The processing industry of dendrobium officinaleis developing rapidly recently,it was called "gold drug" and has the high medicinal value. But the Dendrobium officinale product processing hasn’t been well developed, So it is urgent and important to study the key technologies of processing.In this paper,YuXi dendrobium officinale was the research object. This paper focused on the study of key technologies of processing of the dendrobium officinale oral. Especially optimizing the processing technologies of the extraction processing and clarification processing in this study, and through the anti-oxidation experiment and in vitro digestion experiment to prove the health effects of polysaccharides in dendrobium officinale. The main conclusions were as follows:(1)The extraction technologies:To improve the extraction rate of polysaccharides in dendrobium officinale, it was studied the effect of ultrasound-assisted enzymatic extraction technology and hot water extraction technology. Under the key ultrasound-assisted enzymatic extraction technology the extraction rate of polysaccharides was 13.87%. Under the key hot water extraction technology the extraction rate of polysaccharides was 14.06%.The antioxidant activity and bioavailability of ultrasonic assisted enzymatic polysaccharide was higher than the hot water. Through the comprehensive evaluation of polysaccharide extraction rate, antioxidant activity and bioavailability, the better technology was the ultrasound-assisted enzymatic extraction technology:the PH was 5.5,the time was 105 min, the temperature was 45℃,the Liquid ratio was 1:9, the Ultrasonic power was 150W, the extraction rate of polysaccharides was 13.87%.(2)The clarification technologies:To improve the polysaccharide retention rate and transmittance of dendrobium extracts, it was studied the effect of alcohol precipitation technology and chitosan clarification technology. Under the condition of the optimum alcohol precipitation technology, the oral’s polysaccharide retention was 74.91%, the transmittance was 72.28%; Under the condition of the optimum chitosan clarification technology, the oral’s polysaccharide retention was 83.87%, the transmittance was 80.74%.The antioxidant activity of polysaccharide under chitosan clarification technology was better than the alcohol precipitation technology, And there was no significant difference in the bioavailability of the two technologies. Through the comprehensive evaluation of polysaccharide retention rate and transmittance, antioxidant activity and bioavailability, the better technology was the the chitosan clarification technology:the chitosan usage was 0.05%, the temperature was 50℃, the time was 3 h, the oral’s polysaccharide retention was 83.87%, the transmittance was 80.74%.(3) The formulation technology:using the method of single factor and orthogonal experiments to research best formulation:the β-cyclodextrin was 1.5%,the sucrose was 5.0%, citric acid was 0.13%.(4) Sterilization technology:According to the physical and chemical indicators-. antioxidant activity and bioavailability of oral, the optimum sterilization technology was sterilization by 100℃ boiling water, Sterilization time was 20 min.(5) By accelerating experiments and light accelerating experiments, initially determined the oral liquid should be stored at low temperature in the dark condition.
