Flammuiina velutipes(F.velutipes) is popular in word because of its delicious taste and medicinal value. With the F.velutipes industry continuous development, a large number of F.velutipes waste generation is produced every year and cause a lot of waste. The polysaccharides are very potential precious resources in the root of F.velutipes. Therefore, how to use the F.velutipes waste effectively with modern biological technology is very important recently. However, F. velutipes root polysaccharides (FRP) have rarely been investigated. To find more source of the FRP, an extraction procedure for FRP was developed. The optimizing extraction conditions were chosen by using a response surface design. One fraction (FRP-2) was isolated and purified, and its characteristics were obtained. The study also test the anti-UV activity to C. elegans of FRP, which was to provide theoretical support of F. velutipes polysaccharide in the future.A Box-Benhnken design (BBD) experiment with three factors and three levels, including enzymolysis temperature, microwave time, and microwave power, was designed on the basis of the results of single factor experiments. To improve the polysaccharide yield of F. velutipes root, the following optimal extraction conditions were used:52.67 ℃ enzymolysis temperature,80 s microwave time, and 144 W microwave power. The actual measured value of the yield was 2.06% (w/w) and the predicted value was 2.01% (w/w).The FRP was purified by Sepharose 2B gel column and named the fraction FRP-2. The structural characterization of FRP-2 was carried out by frared spectroscopy (IR), nuclear magnetic resonance spectroscopyand(NMR), high performance liquid chromatography (HPLC) instrument, gaschromatography (GC) and scanning electron microscopy (SEM). The results showed that, the molecular weight was 2.60×105 Da of FRP-2 with many side chain and branch. The monosaccharide composition was glucose mostly and it consisted of pyranose ring with both a-configuration and β-configuration., and most glycosidic bond is 1-4. FRP-2 seemed flocculent at SEM.The C. elegans was taken as the UV model and the UV dosage was chosen 7 mj/cm2. The C. elegans was gaven drug 18 h with the FRP-2 concentrations of 0.05,0.10,0.20, and 0.40 mg/mL. The results showed that, the drug group had higher survival rate and head swing frequency under the FRP-2 concentrations of 0.05-0.20 mg/mL than the blank control group, all the drug groups had higher activity of total superoxide dismutase (T-SOD) and catalase (CAT). The group of 0.10 mg/mL FRP-2 concentrations had higher sod-3 and gst-1 gene expression than the blank control group. All the results showed that the FRP-2 has a certain anti-UV activity on C. elegans.Oatmeal cookies containing FRP were made, which provided reference for the comprehensive development with FRP...
